Off-Label use of Raloxifene hydrochloride in uterine fibroids: A novel insert-based formulation approach and IN-VIVO preclinical evaluation

Summary: Most uterine fibroids treatments are invasive, so using Raloxifene hydrochloride (RLH) as an off-label therapy for uterine fibroids is proposed. An RLH-loaded drug eluting insert (DEI) was formulated and its uterine-targeting efficiency after intrauterine administration was studied. The DEI showed no interference between the drug and excipients, and achieved desired drug release with degradation. Preclinical in vivo results demonstrated reduced size of uterine fibroids with RLH treatment, indicating successful uterine targeting via the vaginal route and potential for reducing side effects.

Design and evaluation of nanostructured lipid carriers loaded with Salvia officinalis extract for Alzheimer's disease treatment

Summary: This study aimed to design, develop, optimize, and characterize freeze-dried Salvia officinalis extract (FSE) loaded nanostructured lipid carriers (NLC) for intranasal administration. NLCs were prepared using a solvent evaporation method and optimized using central composite design (CCD). The optimized formulation showed spherical particles with a smooth exterior and a particle size ranging from 127 +/- 0.68 nm to 140 +/- 0.74 nm. The NLCs exhibited high drug loading and prolonged and controlled release. Protein adsorption studies indicated lower adsorption onto NLCp and higher adsorption onto NLCc surfaces. The modified ORAC assay demonstrated higher antioxidative activity for NLCo and NLCc compared to FSE. Cell culture tests suggested the potential of these NLCs for targeted drug delivery and controlled release in the brain.

Formulation and Characterization of Febuxostat Loaded Nanostructured Lipid Carriers (NLCs)-Gel for Topical Treatment of Gout

Summary: This study aimed to prepare and characterize Nano-Structured Lipid Carriers (NLCs) of Febuxostat (FB) incorporated in a gel for the treatment of Gout. The hot high-pressure homogenization technique was used to improve the physicochemical property of FB. Results showed that NLCs (1:3) had high entrapment and drug content, with 87% of the drug released in a controlled manner within 6 hours. This indicates that the NLCs gel formulation can modify drug release, increase bioavailability, and reduce side effects.

The results showed that the F10 formulation combined with the Air jet nebulizer exhibited the best performance in terms of particle size and drug deposition, making it the most suitable combination for delivering BDP-NLC formulations.

Appraisal of Nano-lipidic Astaxanthin Cum Thermoreversible Gel and its Efficacy in Haloperidol Induced Parkinsonism
