Antileishmanial activities of three chalcone derivatives and their association with plasma membrane rigidity as assessed by EPR spectroscopy

Chalcones: Promising therapeutic agents targeting key players and signaling pathways regulating the hallmarks of cancer

Summary: There is an urgent need for innovative anticancer treatments that are highly effective and have low toxicity due to the development of drug resistant malignancies and the lack of specificity in current treatments. Chalcones, which are the precursors for flavonoids and isoflavonoids, are readily available and have simple chemical structures, allowing for the synthesis of several chalcone derivatives. Both naturally occurring and synthetic chalcone derivatives have shown promising biological activities against various cancer hallmarks, and this review will discuss their mechanisms of action in detail.

Antileishmanial activity of 3,4,5-trisubstituted isoxazoles by interaction with Leishmania amazonensis plasma membrane

Summary: The isoxazole core is essential in discovering new and potent leishmanicidal agents. In this study, 35 3,4,5-trisubstituted isoxazole derivatives were tested against L. amazonensis, with 8 compounds showing potent antileishmanial activity, particularly compounds 2e and 3de which also exhibited low toxicity for mammalian cells.

Ivermectin and curcumin cause plasma membrane rigidity in Leishmania amazonensis due to oxidative stress

Summary: Spin label electron paramagnetic resonance (EPR) spectroscopy was used to study the mechanisms of action of ivermectin and curcumin against Leishmania (L.) amazonensis promastigotes. Both compounds were found to result in increased plasma membrane rigidity due to oxidative processes. Ivermectin showed higher activity at low cell concentrations, but no significant difference was observed at concentrations greater than 1.5 x 108 parasites/mL.

Membrane dynamics in Leishmania amazonensis and antileishmanial activities of β-carboline derivatives

Summary: The beta-carboline compounds 8i and 6d showed antileishmanial activity and selectivity, with lower affinity for the parasite membrane. These compounds caused membrane rigidity in Leishmania promastigotes, leading to modifications in cell structure and reduced parasite numbers in infected macrophages. Moreover, they exhibited greater toxicity towards the protozoa than macrophages, inducing various cellular changes and alterations in the cell cycle.

Identification of Chalcone Derivatives as Inhibitors of Leishmania infantum Arginase and Promising Antileishmanial Agents
