Design, synthesis, biological evaluation, and docking study of chromone-based phenylhydrazone and benzoylhydrazone derivatives as antidiabetic agents targeting α -glucosidase

Summary: In order to develop novel alpha-glucosidase inhibitors, a series of chromone-based phenylhydrazone and benzoylhydrazone derivatives were designed, synthesized, and evaluated for their inhibitory effects on alpha-glucosidase. The compounds showed varying degrees of inhibitory activity, with compound 5c being the most potent inhibitor. Mechanistic studies were conducted to elucidate the inhibitory mechanism of compound 5c on alpha-glucosidase. In addition, in vivo tests showed that compound 5c could suppress the rise of blood glucose levels after sucrose loading in mice, and cytotoxicity assays indicated low cytotoxicity of compound 5c.

Two New Chromones from Cassia auriculata and Their Antiviral Activity

Summary: Two new chromones, 2-(2-(furan-2-yl)-2-oxoethyl)-6-methoxy-8-(2-oxopropyl)-chromone (1) and 2-(2-(furan-2-yl)-2-oxoethyl)-5-methoxy-8-(2-oxopropyl)-chromone (2), along with four known ones (3-6) were isolated from the stem bark of Cassia auriculata. The new compounds were characterized by spectroscopic methods, while the known compounds were identified through comparison with literature data. Compounds 1 and 2 exhibited potential anti-tobacco mosaic virus (anti-TMV) activities with inhibition rates of 28.2% and 30.5% respectively, similar to the positive control. They also showed potential anti-rotavirus activities with therapeutic index (TI) values of 14.8 and 15.9 respectively, lower than the positive control.

Development of novel chromones as antioxidant COX2 inhibitors: in vitro, QSAR, DFT, molecular docking, and molecular dynamics studies

Summary: This article investigates the antioxidant and anti-inflammatory activities of 15 novel chromone derivatives. The antioxidant activity of these compounds was evaluated using a spectrophotometric method and the mechanism of action was discussed using Density functional theory calculations. Molecular docking and dynamics simulations were performed to study the compounds' interaction with the COX2 enzyme. The results show that compounds B3 and B8 exhibit both strong antioxidant activity and COX2 inhibition.

Chemical reactivity of 3-substituted-6,8-dimethylchromones towards 1H-benzimidazol-2-ylacetonitrile and 5-amino-2,4-dihydro-3H-pyrazol-3-one: Spectroscopic, theoretical and in silico ADME studies

Summary: The reactivity of 1H-benzimidazol-2-ylacetonitrile and 5-amino-2,4-dihydro-3H-pyrazol-3-one towards 3-substituted-6,8-dimethylchromones was studied. Different chemical transformations were used to efficiently synthesize various pyrido[1,2-a]benzimidazoles, chromeno[2,3-b]pyridines, pyrazolo[3,4-b]pyridines and chromenopyrazolopyridines. The molecular geometry, reactivity descriptors, and NMR chemical shift values of the synthesized compounds were determined using density functional theory. The compounds also exhibited promising nonlinear optical properties and in silico ADME calculations were performed.

Observations into the reactivity, docking, DFT, and MD simulations of fludarabine and clofarabine in various solvents

Summary: Fludarabine (FA) and Clofarabine (FCA), two novel drugs, were studied both experimentally and theoretically. The solvation free energies of FA and FCA were calculated using different solvents, and their absorption properties were evaluated. Structural features of the drugs were characterized using spectroscopic techniques. Theoretical calculations, molecular docking analysis, and MD simulations were performed to investigate the chemical and electronic characteristics, binding affinities, and stability of the drugs.
