Understanding the screening effect of aqueous DES on the IDPs: A molecular dynamics simulation study using amyloid β42 monomer

Summary: Deep eutectic solvents (DESs) show potential as a replacement for ionic liquids in bio-compatibility solvent engineering. This study investigates the effect of aqueous DESs on the conformation of intrinsically disordered proteins (IDPs), specifically the amyloid beta (A beta(42)) monomer. Molecular dynamics simulations reveal that DES effectively screens the interaction between A beta monomer and water, modulating local protein-solvent interactions. The aqueous DES medium also preserves the secondary structure of A beta(42) monomer through increased intramolecular hydrogen bonding and salt-bridge interactions. This research provides insights into how DES stabilizes an IDP at the molecular level and suggests hindered aggregation of amyloid beta structures in DES medium compared to pure water.
