Polyethylene glycol induced structural modulation of holo alpha-lactalbumin: In vitro and in vivo approach towards protein stability

Molecular basis of structural stability of Irisin: A combined molecular dynamics simulation and in vitro studies for Urea-induced denaturation

Summary: In this study, urea-induced denaturation was used to investigate the conformational changes of irisin. The results showed that the unfolding of irisin occurs in a biphasic manner, with an intermediate state (I) populated at 2.5 M urea concentration. Counteraction of the cetyltrimethylammonium bromide-induced protein aggregation by heparin: Potential impact on protein aggregation and neurodegenerative diseases using biophysical approaches

Summary: This study investigates the effect of heparin on CTAB-induced aggregation of bovine serum albumin (BSA) using biophysical techniques. The results show that heparin completely inhibits CTAB-induced protein aggregation and reduces the apparent rate constant. CTAB acts as a protein-aggregating agent, but heparin prevents the perturbation of secondary and tertiary structure induced by CTAB and the formation of amyloid-like structures. ITC measurements reveal that CTAB and heparin form a complex that does not bind with protein. The CTAB-heparin complex may play a role in inhibiting protein aggregation and could serve as a basis for developing therapeutic strategies for various neurodegenerative complications.

In this review article, the various intermediate states observed and characterized in vitro conditions are discussed, along with their role in regulating the biological function of cells.

Measuring Structural Changes in Cytochrome c under Crowded Conditions Using In Vitro and In Silico Approaches

Summary: It is known that macromolecular crowding in the cell affects protein structure, stability, and function, but predictive studies are still relatively unexplored. This study investigated the effects of polyethylene glycol (PEG 400 Da), ethylene glycol (EG), and their mixture on the structural stability of cytochrome c (cyt c). The results showed that EG stabilizes the protein and has no significant effect on the tertiary structure, while PEG 400 destabilizes the protein structure and stability. However, EG counteracts the destabilizing effect of PEG 400. This study highlights the importance of protein folding and interactions in the crowded cellular environment.

Understanding the nano colloid-protein interaction in crowded milieu

Soft interactions versus hard core repulsions: A journey of cytochrome c from acid-induced denaturation to native protein via pre-molten globule and molten globule conformations exploiting dextran and its monomer glucose
