Relocalization of Translation Termination and Ribosome Recycling Factors to Stress Granules Coincides with Elevated Stop-Codon Readthrough and Reinitiation Rates upon Oxidative Stress

Desislava L. Makeeva et al.

Summary: Upon oxidative stress, mammalian cells reprogram their translation and form stress granules (SGs). Arsenite-induced stress increases stop-codon readthrough rate and translation reinitiation levels on uORF-containing and bicistronic mRNAs. Translation termination factors, ribosome recycling, and translation reinitiation factors are recruited to SGs during arsenite treatment. This spatial redistribution of translation factors may contribute to mRNA translation resumption and suggest the presence of post-termination, recycling, or reinitiation complexes in SGs.

Identification and functional characterization of mRNAs that exhibit stop codon readthrough in Arabidopsis thaliana

Sarthak Sahoo et al.

Summary: This study investigates stop codon readthrough (SCR) in plants and its role in protein localization and function. Analyzing ribosome profiling datasets, the researchers identified 144 genes in Arabidopsis thaliana that exhibit SCR. They found that SCR generates evolutionarily conserved signals and regions in the C-terminal extensions of these proteins, and these genes are involved in translation, photosynthesis, and abiotic stress tolerance. Experimental assays confirmed SCR in representative genes, and microscopy showed the nuclear localization of a SCR product. This research highlights the importance of SCR in plant physiology.

Codon usage and protein length-dependent feedback from translation elongation regulates translation initiation and elongation speed

Xueliang Lyu et al.

Summary: Codon usage bias plays a crucial role in determining gene expression levels and ribosome flux, with an impact on both protein abundance and translation initiation efficiency. Optimal codon usage facilitates production of large proteins, while non-optimal codon usage in long CDS mRNAs inhibits translation initiation by inducing phosphorylation of eIF2 alpha. Additionally, a feedback mechanism from translation elongation, influenced by codon usage and CDS length, regulates translation initiation and elongation kinetics.
