m6A demethylase ALKBH5 attenuates doxorubicin-induced cardiotoxicity via posttranscriptional stabilization of Rasal3

Ri-Feng Gao et al.

Summary: The roles of m(6)A and m(6)A demethylase ALKBH5 in DOX-induced cardiotoxicity (DIC) were investigated. The results showed that ALKBH5 deficiency or overexpression can affect cardiac function and signal transduction mediated by DOX, thereby improving or aggravating DIC injury.

Mitochondrial calpain-1 activates NLRP3 inflammasome by cleaving ATP5A1 and inducing mitochondrial ROS in CVB3-induced myocarditis

Xiaoxiao Liu et al.

Summary: This study found that calpain plays an important role in Coxsackievirus-induced myocarditis and revealed its underlying mechanism of action. The NLRP3 inflammasome was also found to be involved in the process. These findings suggest that calpain may be a promising target for the treatment of viral myocarditis.

Myeloid cell-specific deletion of Capns1 prevents macrophage polarization toward the M1 phenotype and reduces interstitial lung disease in the bleomycin model of systemic sclerosis

Li Zhang et al.

Summary: The study demonstrates that myeloid cell calpain may promote macrophage M1 polarization and pro-inflammatory responses in SSc-related ILD, potentially through the PI3K/AKT1 signaling pathway. Myeloid cell calpain could be a potential therapeutic target for SSc-related ILD in the bleomycin model.

The covalent NLRP3-inflammasome inhibitor Oridonin relieves myocardial infarction induced myocardial fibrosis and cardiac remodeling in mice

Ri-Feng Gao et al.

Summary: Oridonin, a covalent NLRP3 inhibitor, reduces myocardial fibrosis and preserves cardiac function in a mouse MI model, indicating its potential therapeutic effect on acute MI patients. Additionally, Oridonin decreased the expression levels of inflammatory factors and reduced myocardial macrophage and neutrophil influxes in treated mice.

IP3R1 regulates Ca2+ transport and pyroptosis through the NLRP3/Caspase-1 pathway in myocardial ischemia/reperfusion injury

Guixi Mo et al.

Summary: This study revealed the important role of IP3R1 in myocardial ischemia/reperfusion by regulating Ca2+ levels, inflammation, and pyroptosis to improve injury. The binding of ERP44 to IP3R1 can inhibit Ca2+ overload, alleviating pyroptosis and myocardial ischemia/reperfusion injury.

Interaction between extracellular ATP5A1 and LPS alleviates LPS-induced neuroinflammation in mice

Lingyun Yue et al.

Summary: Neuroinflammation is a major factor in Alzheimer's disease, with the presence of LPS in senile plaques indicating its role in AD pathology. The protein ATP5A1 has been found to interact with LPS and alleviate LPS-induced symptoms in a mouse model of neuroinflammation, showing potential in regulating LPS-mediated neuroinflammation in AD.

M2 macrophages, but not M1 macrophages, support megakaryopoiesis by upregulating PI3K-AKT pathway activity

Hong-Yan Zhao et al.

Summary: The study revealed the critical role of macrophages in regulating megakaryocyte production, with M1 and M2 macrophages exhibiting opposing effects in a manner dependent on the PI3K-AKT pathway. This sheds light on the pathogenesis of thrombocytopenia and presents a potential therapeutic strategy for promoting megakaryopoiesis.

Cardiac fibrosis

Nikolaos G. Frangogiannis

Summary: Myocardial fibrosis, characterized by the expansion of cardiac interstitium through deposition of extracellular matrix proteins, is a common pathophysiologic feature in various myocardial conditions. Activated fibroblasts and myofibroblasts play a central role in cardiac fibrosis, producing matrix proteins and triggering fibrogenic signalling cascades in response to stress. Immune cells, vascular cells, and cardiomyocytes can also contribute to fibrosis, while fibrotic changes may disrupt cardiac function and play a role in arrhythmogenesis.
