Summary: This study reveals the mechanism of selectivity in coupling between serotonin receptors and G proteins. It is found that the length of TM5 and TM6 can act as a macro-switch to determine the receptor's preference for G(s) or G(i) proteins. Additionally, specific residues within TM5 and TM6 function as micro-switches to form specific interactions with G(s) or G(i). These findings extend our understanding of ligand recognition at serotonin receptors.

Summary: Despite being expressed in various tissues, the activity of Cdk5 is limited to the central nervous system. Cdk5 plays a crucial role in the pathogenesis of Alzheimer's disease by being abnormally overactivated, leading to the hyper-phosphorylation of ll-amyloid precursor protein and tau and contributing to the development of the disease. Inhibiting the activity of Cdk5-p25 could be a promising strategy for anti-AD therapeutics.
