Synthesis of novel glutarimide ligands for the E3 ligase substrate receptor Cereblon (CRBN): Investigation of their binding mode and antiproliferative effects against myeloma cell lines

Summary: In order to enhance the chemical toolkit for targeted protein degradation, the authors developed a new series of non-thalidomide Cereblon (CRBN) ligands. By using a thio-Michael addition reaction, readily available 2-methylidene glutarimide was converted to a series of 2-((hetero)aryl(methyl))thio glutarimides. These compounds were evaluated for their affinity to human CRBN and their binding modes were studied through X-ray crystallography. The newly identified Cereblon ligands show promising potential for the synthesis of novel PROTAC protein degraders.

Summary: The binucleophilic properties of 1,2-aminothiol and its rare occurrence in nature make it a useful reporter for tracking molecules in living systems. In this study, we expand the chemical space of heteroaromatic nitriles for bioconjugation under physiologically relevant conditions. We provide highly selective and moderately reactive nitriles as well as highly reactive yet less selective analogs with a variety of enabling attachment chemistries to aid future applications in bioconjugation, chemical biology, and nanomaterial science.

Development of Substituted Phenyl Dihydrouracil as the Novel Achiral Cereblon Ligands for Targeted Protein Degradation

Summary: Substituted achiral phenyl dihydrouracil (PDHU) was developed as a novel class of CRBN ligands for PROTACs. Some substituted PDHUs showed comparable binding affinity to lenalidomide. Structural modeling provided insights into the molecular interactions between PDHU ligands and CRBN. PDHUs also demonstrated greater stability than lenalidomide. Finally, potent BRD4 degraders were developed using trisubstituted PDHUs.

Cereblon neo-substrate binding mimics the recognition of the cyclic imide degron

Summary: In targeted protein degradation, immunomodulatory drugs (IMiDs) or cereblon (CRBN) E3 ligase modulatory drugs (CELMoDs) recruit neo-substrate proteins to CRBN for degradation. The recognition mode of natural CRBN degron is described, and it is found that the binding of neo-substrates closely mimics the recognition of natural degrons. The structural basis for the rational design of CRBN effectors is completed.

Development of PDE6D and CK1α Degraders through Chemical Derivatization of FPFT-2216

Summary: Immunomodulatory drugs approved for multiple myeloma treatment exert effects by inducing interactions between the CRL4(CRBN) E3 ubiquitin ligase and a zinc finger degron motif, degrading specific targets. FPFT-2216 compound was shown to degrade PDE6D, IKZF1, IKZF3, and CK1a, leading to the development of more selective degraders TMX-4100 and TMX-4116 targeting PDE6D and CK1a. PDE6D depletion by FPFT-2216 does not impede the growth of KRAS-dependent cells, illustrating challenges in drugging PDE6D-KRAS.

Both PROTAC series demonstrated selective degradation of HDAC6 in leukemia cell lines.

The E3 ligase adapter cereblon targets the C-terminal cyclic imide degron

Preclinical evaluation of proteolytic targeting of LCK as a therapeutic approach in T cell acute lymphoblastic leukemia

Summary: T cell acute lymphoblastic leukemia (T-ALL) is an aggressive hematological malignancy. The LCK inhibitor dasatinib has shown efficacy against T-ALL, but its effect is temporary. Using the PROTAC approach, researchers developed a series of LCK degraders based on dasatinib, with lead compound SJ11646 exhibiting significant efficiency in cereblon-mediated LCK degradation. SJ11646 showed higher cytotoxicity and prolonged suppression of LCK signaling compared to dasatinib, both in vitro and in vivo. SJ11646 also showed binding affinity to several human kinases, making it a potential therapeutic agent for other cancers.

E3 ligase ligand chemistries: from building blocks to protein degraders

Expanding the range of E3 ligands could contribute to broadening the scope of targeted protein degradation. This review focuses on the chemistry aspect of PROTAC design and serves as a blueprint for future research in this complex field.
