DPP9 Comes of Age: Highly Selective Inhibitors Promise New Therapeutic Opportunities

Veronica A. Jimenez

Summary: Dipeptidyl peptidase-like protein 9 (DPP9) is a promising drug target for hematological diseases. Two novel DPP9 inhibitors with nanomolar affinity and unprecedented selectivity over DPP8 have been discovered, opening the way for future DPP9-mediated treatments.

CCR2 and DPP9 expression in the peripheral blood of COVID-19 patients: Influences of the disease severity and gender

Mohsen Sharif-zak et al.

Summary: The gene expression levels of CCR2 and DPP9 vary in COVID-19 patients with different disease forms, with significant differences between male and female patients. Consideration of gender and disease severity is recommended for COVID-19 management.

Vildagliptin-Derived Dipeptidyl Peptidase 9 (DPP9) Inhibitors: Identification of a DPP8/9-Specific Lead

Siham Benramdane et al.

Summary: This study aimed to develop selective DPP8/9 inhibitors using Vildagliptin as a starting point and reducing its affinity for DPP4. Various derivatives were explored, resulting in the discovery of molecules with higher DPP8/9 potency and better DPP9 selectivity.

A Phenotypic Screen Identifies Potent DPP9 Inhibitors Capable of Killing HIV-1 Infected Cells

Keith P. Moore et al.

Summary: Although current antiretroviral therapy can control HIV-1 replication, finding a cure remains a challenge. This study identified a novel chemical class that can kill HIV-1 infected cells by modulating DPP9 and activating CARD8 inflammasome. The compounds ICeD-1 and ICeD-2 were effective in combination with efavirenz, lowering the concentration needed for cell kill without complete DPP9 inhibition.

New insights into the role of dipeptidyl peptidase 8 and dipeptidyl peptidase 9 and their inhibitors

Chenkai Cui et al.

Summary: DPP8/9, as important enzymes widely expressed in mammals, play key regulatory roles in various physiological and pathological processes. Research on DPP8/9 has shown their involvement in multiple biological processes and their potential as therapeutic targets for related diseases.
