Functional analysis of three new alpha-thalassemia deletions involving MCS-R2 reveals the presence of an additional enhancer element in the 5' boundary region

Serena Capasso et al.

Summary: We identified three novel deletions involving the Multispecies Conserved Sequences (MCS) R2 in patients with alpha-thalassemia, and conducted transcriptional and expression analysis to understand the role of different segments of the MCS-R2 element. Our findings suggest the existence of an enhancer element in the region that helps increase the expression of the alpha-globin genes.

Comprehensive Analysis of Congenital Adrenal Hyperplasia Using Long-Read Sequencing

Yingdi Liu et al.

Summary: This study developed a comprehensive analysis method for congenital adrenal hyperplasia (CACAH) using long-read sequencing, which accurately identified pathogenic variants in CAH candidate genes. Compared to traditional methods, CACAH showed higher sensitivity and specificity, providing a comprehensive approach for CAH genetic diagnosis and carrier screening.

Thalassaemia

Antonis Kattamis et al.

Summary: Thalassaemia is a genetic disorder that affects globin chain synthesis and has a worldwide distribution. The unbalanced production of globin chains leads to ineffective erythropoiesis, increased haemolysis, and disrupted iron homeostasis. The clinical phenotype varies from mild to severe, with treatment options including transfusion, iron chelation, allogeneic haematopoietic stem cell transplantation, and gene therapy.

Comprehensive Analysis of Fragile X Syndrome: Full Characterization of the FMR1 Locus by Long-Read Sequencing

Qiaowei Liang et al.

Summary: This study developed a new genetic testing method, comprehensive analysis of FXS (CAFXS), which utilizes long-range PCR and long-read sequencing to accurately detect FMR1 CGG expansions and identify AGG interruptions, rare intragenic variants, and large gene deletions. CAFXS greatly improves the accuracy of genetic screening and diagnosis for fragile X syndrome.

Detection of four rare thalassemia variants using Single-molecule realtime sequencing

Shiqiang Luo et al.

Summary: This study utilized single molecule real-time technology (SMRT) sequencing to identify four rare variants of thalassemia that had not been detected by conventional diagnostic methods. The researchers also conducted genotype and phenotype analyses on family members of thalassemia patients. The findings demonstrated the significant application value of SMRT in diagnosing rare alpha-globin and ss-globin gene cluster deletions, gene recombination, and deletion breakpoints in thalassemia.

The Evolving Role of Next-Generation Sequencing in Screening and Diagnosis of Hemoglobinopathies

Ahlem Achour et al.

Summary: Next-generation sequencing (NGS) has rapidly transitioned from a research setting to a clinical application, becoming the preferred method for detecting disease-causing variants in various genetic diseases. NGS plays an important role in screening and diagnosing hemoglobinopathies, with added value in large-scale screening and complex cases. It is a useful addition to existing methods for diagnosing these disorders.
