Focusing on a nickel hydrocorphinoid in a protein matrix: methane generation by methyl-coenzyme M reductase with F430 cofactor and its models

Summary: Methyl-coenzyme M reductase (MCR) is an important enzyme that catalyzes anaerobic methane generation and oxidation. Extensive studies have been conducted on the substrate-binding cavity and the cofactor F430 in MCR, revealing two different reaction mechanisms. In addition to native MCR, model complexes and protein-based functional models have been used to investigate the functionality of MCR. These studies contribute to understanding the enzymatic reaction and have important implications for replicating the methane-generation process.

Reactivity of Myoglobin Reconstituted with Cobalt Corrole toward Hydrogen Peroxide

Summary: The incorporation of a cobalt complex of corrole (CoCor) into an apo-form of myoglobin to provide a reconstituted protein (rMb(CoCor)) was characterized by UV-vis, EPR, and mass spectroscopic measurements. Reaction of rMb(CoCor) with hydrogen peroxide leads to irreversible oxidation of the CoCor cofactor, while the same reaction in the presence of a phenol derivative yields the cation radical form of CoCor. Detailed kinetic investigations suggest the formation of a transient hydroperoxo complex of rMb(CoCor) which promotes the oxidation of phenol derivatives.

Ratiometric Oxygen Sensing with H-NOX Protein Conjugates

Summary: Ratiometric sensors are a type of self-referencing constructs that can function in cells and tissues, and their read-out is not affected by sensor concentration. This study developed protein-based ratiometric oxygen sensors using heme nitric oxide/oxygen-binding protein (H-NOX) and oxygen-responsive fluorescent probes. By modifying the amino acid residues on the protein surface, the oxygen sensitivity of the sensor can be adjusted.

Syntheses and Properties of Metalated Tetradehydrocorrins

Summary: This study reports the synthesis and characterization of a new type of metal complex using crystallographic, electrochemical, computational, and spectroscopic techniques. The pyrene moiety increased interactions with both HSA and CT-DNA, likely due to pyrene planarity, while the fluorinated corroles showed different interaction modes with HSA and CT-DNA.
