Optimized flow cytometry assays to monitor neutrophil activation in human and mouse whole blood samples

Summary: In this study, researchers developed simple assays to determine the activation state of polymorphonuclear neutrophils (PMNs) in human and mouse blood samples. By measuring reactive oxygen species production and the expression of cell surface markers, they were able to accurately assess PMN functions. ATP breakdown in plasma of children limits the antimicrobial effectiveness of their neutrophils

Summary: We investigated the changes in ATP and ADO levels in the plasma of children with age and their influence on the antimicrobial efficacy of their PMNs. We found that plasma ATP levels correlated with age and were significantly lower in infants than in adolescents. In addition, the activities of the enzymes responsible for ATP and ADO breakdown were higher in children than in adolescents and correlated inversely with age and antimicrobial capacity of PMNs. Similarly, young mice had lower plasma ATP levels and weaker bacterial phagocytosis by PMNs compared to adolescent mice. However, ATP supplementation could recover the bacterial phagocytosis of young mice to levels similar to those of adolescent mice.

High levels of extracellular ATP lead to different inflammatory responses in COVID-19 patients according to the severity

These findings provide valuable insights into the changes in purinergic signaling and immune response in COVID-19, highlighting the therapeutic potential of the purinergic system as a target for the treatment of SARS-CoV-2 disease.
