AnAzf1 acts as a positive regulator of ochratoxin A biosynthesis in Aspergillus niger

Summary: Aspergillus niger produces ochratoxin A (OTA), which is genotoxic and carcinogenic. In this study, a gene encoding the transcription factor Azf1 was deleted in A. niger, resulting in complete blockage of OTA production. The deletion of Azf1 also led to decreased levels of reactive oxygen species (ROS) and impaired oxidative phosphorylation, which ultimately prevented OTA biosynthesis.

Summary: This study evaluated the impact of water activity and temperature on the growth, gene expression, and aflatoxin production of Aspergillus flavus in Senna. The findings showed that the peak expression of regulatory genes and the maximum toxin production occurred at 28 degrees C and 0.96 water activity. Low water activity and low temperature were identified as ideal conditions to prevent toxin production.

Systematic Characterization of bZIP Transcription Factors Required for Development and Aflatoxin Generation by High-Throughput Gene Knockout in Aspergillus flavus

Summary: The bZIP transcription factor plays a crucial role in fungal development, nutrient utilization, secondary metabolite biosynthesis, and stress response. This study focused on the role of bZIP genes in A. flavus, a major aflatoxin-producing fungus. By using a gene knockout method, 15 bZIP genes were deleted and their functions were analyzed. The results showed that these genes are involved in various physiological processes, including mycelial growth, conidiation, aflatoxin biosynthesis, stress response, and virulence. The findings provide comprehensive insights into the role of bZIP transcription factors in this globally important aflatoxigenic fungus.

Cysteine Inhibits the Growth of Fusarium oxysporum and Promotes T-2 Toxin Synthesis through the Gtr/Tap42 Pathway

Additionally, two novel regulators (AFLA_085880 and AFLA_015850) were identified as critical positive regulators for AFLA synthesis, with their mutants showing decreased expression of AFs cluster genes and subsequent AFB(1) biosynthesis.
