Combinatorial physical methods for cellular therapy: Towards the future of cellular analysis?

Summary: The physical energy activated techniques for cellular delivery and analysis is a rapidly expanding research area in biological and biomedical studies. These techniques have been widely used for delivering biomolecules into different cell types, but individual methods have limitations. Recent studies have combined different membrane disruption techniques, resulting in enhanced transfection, lower delivery parameters, and high cell viability. These studies open up new avenues for understanding subcellular structure and developing novel therapeutic strategies.

Summary: Genetically engineered CAR T-cell therapy makes use of the immune system to eliminate tumors and redirect cytotoxic functions towards cells expressing specific tumor-restricted antigens. Despite FDA approval for 6 CAR T-cell therapies in hematological malignancies, there are limitations in the treatment of both liquid and solid tumors involving T cell-intrinsic, T cell-extrinsic, and therapeutic factors. This review discusses the core structure of CAR, the evolution of different CAR generations, limitations of CAR T-cell therapy, and current strategies to overcome barriers for successful therapy.

Cross-platform immunophenotyping of human peripheral blood mononuclear cells with four high-dimensional flow cytometry panels

Summary: This article highlights the importance of immunophenotyping using high dimensional flow cytometry and presents four high parameter flow cytometry panels for deep immunophenotyping of human PBMC. These panels include 64 cell surface markers and provide insights into immune compartment populations. Adoptive Cellular Therapy with Autologous Tumor-Infiltrating Lymphocytes and T-cell Receptor- Engineered T Cells Targeting Common p53 Neoantigens in Human Solid Tumors

Summary: Adoptive cellular therapy (ACT) targeting TP53 mutations shared among different cancer types shows durable clinical responses, but limited efficacy. A library of 39 T-cell receptors (TCRs) targeting shared TP53 mutations in 7.3% of solid tumor patients was reported. Treatment with ex vivo-expanded autologous tumor-infiltrating lymphocytes (TILs) or autologous peripheral blood lymphocytes transduced with an allogeneic HLA-A*02-restricted TCR specific for p53R175H showed improved efficacy.
