Tingible body macrophages arise from lymph node-resident precursors and uptake B cells by dendrites

Neta Gurwicz et al.

Summary: This study reveals that Tingible body macrophages (TBMs) originate from lymph node-resident precursors and consume germinal center B cells through dynamic protrusions to prevent intracellular-content dissemination. The dynamics and function of TBMs provide insights into the emergence of various antibody-mediated autoimmune conditions.

Heparan sulfate regulates IL-21 bioavailability and signal strength that control germinal center B cell selection and differentiation

Zhian Chen et al.

Summary: In antibody responses, mutated germinal center B (BGc) cells are positively selected for reentry or differentiation, with the support of TFH cell-derived signals including CD40 and IL-21. The binding and signaling of IL-21 in BGc cells is reduced compared to non-BGc cells, due to low cellular heparan sulfate (HS) sulfation. Ndst1-mediated N-sulfation of HS in B cells promotes IL-21 binding and signal strength, and selective desensitization to IL-21 occurs in BGc cells. Therefore, the biochemical regulation of IL-21 availability and signal strength plays a crucial role in GC selection.

Germinal Centers

Gabriel D. Victora et al.

Summary: This paper reviews recent developments in the field of germinal center (GC) biology, specifically in GCs induced by infection or immunization. It summarizes the phenotype and function of different cell types in the GC, focusing on GC B cells. It also examines the cellular and molecular basis of affinity-dependent selection within the GC and the export of memory and plasma cells. Additionally, it provides an overview of the emerging field of GC clonal dynamics and its impact on the diversity of antibodies secreted into serum.

Targeting BMI-1 in B cells restores effective humoral immune responses and controls chronic viral infection

Andrea Di Pietro et al.

Summary: Research reveals that BMI-1 plays a crucial role in chronic viral infections, and targeting BMI-1 can enhance humoral immune responses and accelerate viral clearance. Deficiency of BMI-1 can increase neutralizing and effector function of antibodies, ultimately restoring antibody quality.

CCND3 is indispensable for the maintenance of B-cell acute lymphoblastic leukemia

Franz Ketzer et al.

Summary: CCND3 is essential for the proliferation and survival of B-ALL, exceeding the expression levels of CCND1 and CCND2 at the mRNA level. FOXO1 is identified as a CCND3-activating transcription factor in B-ALL, playing a crucial role in cell growth and survival. The anti-apoptotic effect of CCND3 in B-ALL is independent of the kinase activity of the CCND3-CDK4/6 complex.

Autoantibodies in Patients With Immune-Related Adverse Events From Checkpoint Inhibitors A Systematic Literature Review

Nilasha Ghosh et al.

Summary: Autoantibodies are sometimes associated with immune-related adverse events (irAEs) from immune checkpoint inhibitors (ICIs), but their presence before ICI initiation and their frequency were not known. This study found that autoantibodies were present in close to 50% of patients with ICI-associated endocrinopathies and more than 50% of patients with skin irAEs. However, the positivity rate of autoantibodies was lower in irAEs affecting other organ systems. There are mixed results regarding the utility of autoantibodies as biomarkers of irAEs.

Competition for refueling rather than cyclic reentry initiation evident in germinal centers

Ziqi Long et al.

Summary: Research has found that cyclic reentry initiation and B cell refueling are independently regulated in germinal centers (GCs). Cyclic reentry initiation is associated with B cell receptor affinity, while B cell refueling is limited by both T cell affinity and cells' intrinsic antigen-acquiring abilities. This less binary selection mechanism supports complex antibody maturation pathways and clonal diversity in GCs.

Interleukin-21, acting beyond the immunological synapse, independently controls T follicular helper and germinal center B cells

Isaak Quast et al.

Summary: Germinal centers (GCs) are transient structures within B cell follicles that require coordinated behavior of T and B cells. IL-21, a cytokine derived from T cells, plays a key role in GC biology through its independent actions on T and B cells. IL-21 promotes the expansion and differentiation of CD4(+) T cells and specifically impacts B cell centroblast identity and plasma cell differentiation within the GC. IL-21's activity is not solely dependent on T-B interactions but also on its own growth-promoting effects.

Cyclin D3 drives inertial cell cycling in dark zone germinal center B cells

Juhee Pae et al.

Summary: During affinity maturation, GC B cells undergo proliferation and hypermutation in the dark zone, followed by selection in the light zone. Cyclin D3 plays a crucial role in controlling the anatomical segregation and cell cycle of GC B cells, and a specific mutation can lead to clonal B cell lymphoproliferation.

Follicular dendritic cells restrict interleukin-4 availability in germinal centers and foster memory B cell generation

Lihui Duan et al.

Summary: The study shows that the cytokine IL-4 plays a critical role in B cell fate decisions within germinal centers, affecting the formation of pre-memory cells. Furthermore, deletion of IL4Ra from FDCs impairs the selection of affinity-matured B cells and the generation of memory cells.

Summary: IL-21 plays a crucial role in promoting cell cycle progression within the germinal centre (GC) light zone, facilitating release from the G1 cell cycle stage. This cytokine is essential for establishing normal zone representation and preventing cell accumulation in the G1 cell cycle stage in the GC LZ, highlighting its importance in antibody affinity maturation.
