Lung fibroblasts facilitate pre-metastatic niche formation by remodeling the local immune microenvironment

Zheng Gong et al.

Summary: This study identified a population of fibroblasts that express COX-2 and remodel the lung immune microenvironment, promoting breast cancer metastasis. Genetic ablation of COX-2 in fibroblasts reversed the immune-suppressive phenotypes and improved the anti-metastatic activity of DC-based therapy and PD-1 blockade.

Tumor-polarized GPX3+ AT2 lung epithelial cells promote premetastatic niche formation

Zixin Wang et al.

Summary: By single-cell sequencing, a tumor-polarized subpopulation of alveolar type 2 (AT2) epithelial cells with increased expression of glutathione peroxidase 3 (GPX3) and high production of interleukin (IL)-10 in the premetastatic niche (PMN) was identified. These IL-10-producing GPX3(+) AT2 cells inhibited CD4(+) T cell proliferation but enhanced regulatory T cell generation, providing new insights into immune evasion in lung metastasis.

Lipid-laden lung mesenchymal cells foster breast cancer metastasis via metabolic reprogramming of tumor cells and natural killer cells

Zheng Gong et al.

Summary: In breast cancer models, lung-resident mesenchymal cells (MCs) accumulate neutral lipids, mediated by hypoxia-inducible lipid droplet-associated (HILPDA). These lipid-laden MCs transport lipids to tumor cells and natural killer (NK) cells via exosome-like vesicles, promoting tumor cell survival and proliferation and impairing NK cell function. Blocking inter-leukin-10 (IL-10) improves the efficacy of adoptive NK cell immunotherapy in mitigating lung metastasis.

Targeting metastatic cancer

Karuna Ganesh et al.

Summary: Improved understanding of metastases biology has led to advancements in targeting and treating both micro- and macrometastases. Recent studies have identified the unique biology of metastasis-initiating cells, providing new opportunities for developing more effective treatment strategies through cancer sequencing, mechanistic studies, and clinical trials. These insights offer potential for better outcomes in targeting metastatic relapse and improving patient survival.

Hepatocellular carcinoma

Josep M. Llovet et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global health challenge with rising incidence projected to exceed 1 million cases by 2025. Infection by hepatitis B and C viruses remains a major risk factor for HCC, while non-alcoholic steatohepatitis is emerging as a more common risk factor in Western countries. Advances in systemic therapies for HCC, including immunotherapies and targeted therapies, are expected to revolutionize the management of this disease.

Blocking IL1 Beta Promotes Tumor Regression and Remodeling of the Myeloid Compartment in a Renal Cell Carcinoma Model: Multidimensional Analyses

David H. Aggen et al.

Summary: The study suggests that intratumoral immunosuppression mediated by myeloid-derived suppressor cells and tumor-associated macrophages may lead to immune checkpoint inhibitor resistance in solid tumors. Blocking IL1 beta in combination with anti-PD-1 or cabozantinib has shown increased antitumor activity by reducing immunosuppressive myeloid cells and increasing M1-like tumor-associated macrophages.
