An overview of polyurethane biomaterials and their use in drug delivery

Biodegradable and antithrombogenic chitosan/elastin blended polyurethane electrospun membrane for vascular tissue integration

Summary: Biodegradable vascular membranes composed of PU, CS, and elastin were fabricated using electrospun nanofibers. The membranes showed increased fiber diameter, pore size, and porosity percentage when CS and elastin were incorporated. The membranes exhibited controlled release of CS and elastin and demonstrated antithrombogenic properties with delayed blood clotting time in comparison to PU membranes.

Stretchable and biodegradable chitosan-polyurethane-cellulose nanofiber composites as anisotropic materials

Summary: In this study, cellulose nanofibers (CNFs) were incorporated in chitosan-polyurethane composites to enhance their stretchability and anisotropy. Biodegradable polyurethane was synthesized and combined with CNFs to form the DP-CNF composite crosslinker (DPF). The DPF crosslinker was then blended with chitosan solution to prepare polyurethane-CNF-chitosan (DPFC) composites. The resulting anisotropic films showed great potential for biomimetic and tissue engineering applications.

Summary: In this study, the biocompatibility and stability of CarboSil were improved by introducing 30% PDMS. Both in cell experiments and animal experiments, CarboSil-30% PDMS showed good results. The modified material exhibited superior performance in promoting degradation and reducing calcium deposition, making it promising for use in cardiovascular device manufacturing.

Recent Advances in Polyurethane/POSS Hybrids for Biomedical Applications

Summary: Advanced organic-inorganic materials, such as composites, nanocomposites, and hybrids, offer unique properties for biomedical applications. Polyhedral oligomeric silsesquioxanes (POSS) are one of the most promising inorganic additives due to their biocompatibility, non-toxicity, and phase separation ability. When incorporated into polyurethane matrices, POSS significantly alter the polymer properties, including mechanical properties, surface characteristics, and bioactivity. This review focuses on the development of POSS-PU composites for various biomedical applications, including scaffolds, cardiovascular devices, bio-imaging, and cancer treatment. Methods of characterization and modification of polyurethane-based materials with silsesquioxanes are also discussed.

Biodegradable polyurethane scaffolds in regenerative medicine: Clinical translation review

Summary: Early explorations of tissue engineering and regenerative medicine commonly used simple polyesters as scaffolds. However, with the advancement of technology, a greater variety of synthetic biomaterials, such as biodegradable polyurethanes, have been employed. These polyurethanes can be customized based on their composition and processability, and can incorporate bioactive agents to achieve desired biological responses. While there is much enthusiasm and growth in the literature on biodegradable PU scaffolds, their regulatory approval and routine clinical use are still limited. Nonetheless, there is a growing body of research on the development and pre-clinical applications of these scaffolds.

Summary: The study demonstrates that scaffold architecture can modulate immune responses to drive BM-MNC engraftment, with higher porosity scaffold enhancing the homing and engraftment of BM-MNCs. Histological analysis shows that HP scaffolds lead to thicker fibrous capsule formation and increased collagen deposition, with more native immune cells present compared to LP scaffolds. Different scaffold structures impact local inflammation and drive differential engraftment and remodeling behavior of infused BM-MNCs.

Degradability of Polyurethanes and Their Blends with Polylactide, Chitosan and Starch

Summary: The study found that PUR samples with a higher amount of R,S-PHB and a lower amount of polycaprolactone triol (PCLtriol) degraded the most, with blending with starch (St) having the greatest impact on degradation susceptibility. Although the rate of weight loss of the samples was low, significant reductions in molecular weight, changes in morphology, and thermal property changes indicated that the samples would degrade quickly after a certain period of time.
