Digital and Analog Detection of SARS-CoV-2 Nucleocapsid Protein via an Upconversion-Linked Immunosorbent Assay

Summary: This article investigates a novel approach using upconversion nanoparticles for the detection of the COVID-19 virus and its SARS-CoV-2 variant. The upconversion-linked immunosorbent assay (ULISA) demonstrated a higher sensitivity than conventional assays, with a limit of detection of 1.3 pg/mL. Furthermore, the digital ULISA improved the detection limit by a factor of 10 compared to analog readout for SARS-CoV-2.

Improvement of ligand-free modification strategy to obtain water-stable up-converting nanoparticles with bright emission and high reaction yield

Summary: Sensitive immunoassays are essential for detecting low-abundance cardiac biomarker troponin in blood. Digital assays using photon-upconversion nanoparticles (UCNP) can detect single biomarker molecules, with the size, brightness, and surface architecture of UCNP labels being more crucial than the detection mode. Both analog and digital detection modes show approximately the same assay sensitivity, reaching a limit of detection of 10 pg mL(-1) in plasma, which is within the range of troponin concentrations in healthy individuals.
