Risk factors associated with prevalent vertebral fractures in Duchenne muscular dystrophy

Kim Phung et al.

Summary: This study found that markers of systemic glucocorticoid exposure, such as shorter stature, greater bone age delay, and lower lumbar spine bone mineral density, were associated with spine fragility in patients with DMD.

Cessation of ambulation results in a dramatic loss of trabecular bone density in boys with Duchenne muscular dystrophy (DMD)

N. J. Crabtree et al.

Summary: Glucocorticoids are used to improve muscle strength and prolong ambulation in boys with DMD, but their effect on bone health remains unclear. This study compared bone strength between healthy children and boys with DMD, finding that all boys with DMD had reduced bone and muscle strength. Boys who lost ambulation had significantly lower muscle function, and after two years, they had 63% less bone strength compared to healthy boys. The most significant difference was in trabecular bone density for boys who became non-ambulant, suggesting accelerated bone density loss in DMD boys who lose independent ambulation.

Effect of Different Corticosteroid Dosing Regimens on Clinical Outcomes in Boys With Duchenne Muscular Dystrophy A Randomized Clinical Trial

Michela Guglieri et al.

Summary: A double-blind, parallel-group randomized clinical trial was conducted to compare the efficacy and adverse effects of different corticosteroid regimens in boys with Duchenne muscular dystrophy. The study found that daily prednisone and daily deflazacort were more effective than intermittent prednisone for improving motor function, pulmonary function, and satisfaction with treatment over a 3-year period. There was no significant difference between the two daily corticosteroid regimens.

Randomized Controlled Trial Evaluating the Use of Zoledronic Acid in Duchenne Muscular Dystrophy

Margaret Zacharin et al.

Summary: The study found that using zoledronic acid in DMD patients significantly improved bone mineral density. Compared to the control group, the ZA group had a significantly higher LS BMD Z-score difference at 12 and 24 months, with no impact on pain, mobility, and bone turnover. Additionally, the ZA group showed improvements in pQCT measurements compared to the control group.

The effect of oral bisphosphonate therapy on vertebral morphometry and fractures in patients with Duchenne muscular dystrophy and glucocorticoid-induced osteoporosis

Nat Nasomyont et al.

Summary: This study evaluated the effects of oral bisphosphonate therapy on vertebral fractures in patients with DMD. The results showed stable prevalence and severity of vertebral fractures over 5 years of treatment, suggesting oral BP may be beneficial for secondary prevention of glucocorticoid-induced osteoporosis in this population.
