Higher Maternal Body Mass Index Is Associated With Lower Placental Expression of EPYC: A Genome-Wide Transcriptomic Study

Multimodal profiling of term human decidua demonstrates immune adaptations with pregravid obesity

Summary: The diversity of leukocytes at the maternal-fetal interface has been well-studied in the first trimester, but the immune landscape of the term decidua is still not well-understood. In this study, we analyzed leukocytes from term decidua collected during scheduled cesarean deliveries. Our findings suggest a shift from NK cells and macrophages to T cells and increased immune activation compared to the first trimester. We also observed diversity within decidual macrophages, which was associated with maternal body mass index. These findings provide insight into potential mechanisms for protecting the fetus against maternal inflammation and can be used for future studies on pathological conditions affecting fetal health and reproductive success.

Local immune recognition of trophoblast in early human pregnancy: controversies and questions

Summary: The role of the maternal immune system in reproductive success in humans is controversial, with focus on the interactions between maternal leukocytes and fetal trophoblast cells in the first few weeks of pregnancy, particularly involving allorecognition by T cells and NK cells. NK cells are dominant in the decidua during the first trimester and are believed to establish a boundary between the mother and fetus through allorecognition. Definitive evidence of allorecognition by decidual T cells during the first trimester is still lacking. It is proposed that while T cell-mediated immune responses towards placental trophoblast are minimized during the crucial period of placenta establishment, NK cell allorecognition contributes to successful implantation and healthy pregnancy. Understanding the interactions between trophoblast cells and maternal uterine immune cells in early pregnancy is key to understanding major human pregnancy disorders arising from inadequate maternal oxygen and nutrient supply.
