Nano-liquid chromatography with monolithic stationary phase based on naphthyl monomer for proteomics analysis

Summary: Monolithic poly(2-vinylnaphthalene-co-divinylbenzene) columns were introduced and evaluated as separation media for nano-liquid chromatography. The columns were prepared through in-situ polymerization and characterized by SEM and chromatographic analyses. The columns showed excellent hydrophobic and pi-pi interactions, achieving high theoretical plates numbers and demonstrating potential in protein separation and peptide analysis.

The benefits of mixed-mode chromatography columns for separation of peptides and protein digests

Summary: This study evaluated and compared the use of mixed-mode chromatography and reversed-phase chromatography for separating peptides and protein digests. The effects of pH and organic modifier on retention, resolution, and peak shape were investigated on various columns. Different types of peptides were selected to describe the chromatographic behavior under different conditions. The results showed that mixed-mode chromatography columns have the potential for analyzing differently charged peptides in a single run. The applicability of the tested conditions was verified using cytochrome C digested fragments.

On-line Sequencing of CRISPR Guide RNAs and Their Impurities via the Use of Immobilized Ribonuclease Cartridges Attached to a 2D/3D-LC-MS System

Summary: This study introduced for the first time the automated digestion and sequencing of an RNA molecule using immobilized RNase cartridges attached to a multidimensional LC-MS system. The on-line approach showed higher efficiency and lower sample requirement compared to the conventional off-line approach. The study also developed a three-dimensional LC-MS method for sequencing fractions collected on-line in order to gain insights into the cause of main peak tailing.
