One-step competitive assay for detection of thrombin via disassembly of diblock oligonucleotide functionalised nanogold aggregates

Yu Huang et al.

Summary: In this study, a simple, sensitive, and cost-effective sensing strategy for thrombin was developed. The strategy involved the use of polyadenine diblock oligonucleotides anchored on gold nanoparticles (AuNPs) to detect thrombin. Binding of thrombin to its aptamer triggered the disassembly of AuNP aggregates, resulting in a significant signal change. The assay demonstrated a detection limit of 0.81 pM and a dynamic range from 10 pM to 10 nM, and could be performed in diluted serum solutions without sophisticated instruments.

Human serum albumin depletion based on dye ligand affinity chromatography via magnetic microcryogels

Merve Asena Ozbek et al.

Summary: One of the primary purposes of proteomic studies is to analyze blood proteins as biomarkers. This study developed an alternative approach for albumin depletion by covalently attaching Cibacron Blue 3GA (CB) to magnetic microcryogels. Experimental studies demonstrated that CB-attached magnetic microcryogels can be reused without loss of performance.

A combined computational and experimental approach predicts thrombin adsorption to zeolites

Jiachen Li et al.

Summary: In this study, a combined computational and experimental approach was used to determine the adsorbed sites and orientations of thrombin binding to Ca2+-exchanged LTA-type (CaA) zeolite. It was found that thrombin undergoes changes in secondary structure after adsorption on CaA zeolite, and there is a preferential interaction between thrombin and CaA zeolite, leading to blockage of some thrombin substrate sites. This study not only provides a promising method for characterizing protein-nanoparticle interaction, but also offers insights into the design and application of zeolite with high procoagulant activity.

Metal-chelated polyamide hollow fiber membranes for ovalbumin purification from egg white

Merve Asena Ozbek et al.

Summary: In this study, the purification of ovalbumin was achieved by modifying polyamide hollow fiber membranes using immobilized metal affinity chromatography technique. The modified membranes were characterized and the parameters affecting the adsorption capacity were investigated. The maximum adsorption capacities and desorption ratios were determined. The purity of ovalbumin was evaluated by fast performance liquid chromatography (FPLC) and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) method.

Interface imprinted polymers with well-oriented recognition sites for selective purification of hemoglobin

Canan Armutcu et al.

Summary: This study introduces a novel strategy involving sacrificial metal oxide particles to design interface imprinted polymers, resulting in membranes with high rebinding capacity and excellent selectivity. The use of zinc oxide particles not only preserves imprint cavities in the polymer network but also enhances template removal and rebinding kinetics, making it a simple and effective method for biomacromolecule separation.
