Protein adsorption to diethylaminoethyl-dextran grafted macroporous cellulose microspheres: A critical pore size for enhanced adsorption capacity and uptake kinetic

Liangzhi Qiao et al.

Summary: In this study, four diethylaminoethyl-dextran grafted macroporous cellulose microspheres with different pore sizes were used to investigate the adsorption equilibria and kinetics of bovine serum albumin (BSA) and gamma-globulin. The results showed that both proteins exhibited increasing-then-decreasing trends with decreasing pore size. Furthermore, a critical pore size (CPZ) was identified at which the adsorption capacity and uptake kinetics were significantly enhanced for both proteins.

Mass transfer of proteins in chromatographic media: Comparison of pure and crude feed solutions

Markus C. Berg et al.

Summary: This study investigates the differences in adsorption and diffusion parameters of basic human fi-broblast factor 2 in a simple and complex feed solution in protein chromatography. The slower mass transfer in the complex feed solution was induced by components of the clarified homogenate, leading to a decrease in mass transfer rate.

Relationship betweenHETPmeasurements and breakthrough curves in short chromatography columns

Joey A. Roberts et al.

Summary: The analysis shows that radial flow nonuniformity has a significant impact on breakthrough curves, while axial dispersion has only a minor effect. Higher velocities at the column wall lead to fronting tracer peaks and broader breakthrough curves, while lower velocities at the wall result in tailing peaks and curves. Experimentally, tracer peaks and protein breakthrough curves in mini-columns are consistent with higher velocities at the wall, indicating the importance of flow uniformity in obtaining meaningful binding capacity measurements.

Recent advances in protein chromatography with polymer-grafted media

Linling Yu et al.

Summary: This review discusses the effectiveness of polymer-grafted media in creating high-capacity and high-uptake rate protein chromatography. The focus is on ion-exchange chromatography behavior and the limitations of polymer-grafted media in hydrophobic interaction chromatography and affinity chromatography. Recent advances in theoretical and experimental investigations on polymer-grafted media are also highlighted.

Effect of pore structure on protein adsorption mechanism on ion exchange media: A preliminary study using low field nuclear magnetic resonance

Chao Chen et al.

Summary: This study investigated the adsorption process of BSA, OVA, and IgG on different media using low field NMR and established a theoretical model to describe the relationship between protein binding amount and T-2 relaxation signals. The results showed that smaller proteins tended to form monolayer adsorption on media without dextran chains, while larger proteins exhibited similar adsorption behavior on all tested media. The study provides insights into the protein adsorption mechanism and the impact of pore size variation.

Patterns of protein adsorption in ion-exchange particles and columns: Evolution of protein concentration profiles during load, hold, and wash steps pre dicte d for pore and solid diffusion mechanisms

Juergen Beck et al.

Summary: Understanding protein transport mechanism in ion exchanges is crucial for modeling separation performance. Different transport mechanisms, such as pore diffusion and solid diffusion, exhibit distinct characteristics at various Langmuir equilibrium constants. During hold and wash steps, protein diffusion plays a significant role in solid diffusion, while protein mobility is considerable at low and moderate binding strengths in pore diffusion.
