A Photoswitchable Ligand Targeting the β1-Adrenoceptor Enables Light-Control of the Cardiac Rhythm

Summary: In this study, a new strategy for light-controlled activation of beta(1)-AR was reported using photoswitchable drugs with high selectivity for beta(1)-/beta(2)-AR. The molecules reported in this study allowed for efficient real-time optical control of receptor function in vitro. The best hit, pAzo-2, showed reversible photocontrol of binding and enabled dynamic cardiac rhythm management on living zebrafish larvae using light.

Optical control of Class A G protein-coupled receptors with photoswitchable ligands

Summary: The field of photopharmacology for Class A GPCR ligands has gained attention. This review analyzes 31 papers on photoswitchable ligands for Class A GPCRs and identifies the overarching goal of this research area to be the control of receptor activity with light. The design, photochemistry, and pharmacology of photoswitchable ligands are analyzed, and the trends, challenges, and limitations are discussed. The review presents efficient photoswitchable ligands capable of optically modulating GPCR function in various in vitro assays, as well as highlights the first reports of optical modulation of in vivo GPCR function.

Hypothesis-Driven, Structure-Based Design in Photopharmacology: The Case of eDHFR Inhibitors

Summary: Photopharmacology uses light to regulate the biological activity of drugs. Computer-aided drug design is an attractive approach for designing photoswitchable drugs. In this study, different structure-based approaches for photopharmacology were evaluated using Escherichia coli dihydrofolate reductase (eDHFR) as a case study. Through iterative examination, azobenzene-based eDHFR inhibitors with the thermally metastable cis-isomer emerged as the most promising design strategy. Three inhibitors that could be activated upon irradiation were identified. This systematic study provided valuable insights for the rational design of photoswitchable drugs in the future.

Optical control of the β2-adrenergic receptor with opto-prop-2: A cis-active azobenzene analog of propranolol
