Sodium butyrate supresses malignant human mast cell proliferation, downregulates expression of KIT and promotes differentiation

Clayton A. MacDonald et al.

Summary: Sodium butyrate (NaBu), a histone deacetylase inhibitor (HDACi), inhibits the proliferation and metabolic activity of transformed human mast cell lines without affecting their viability. NaBu significantly blocks the cell cycle progression of HMC-1.1 and HMC-1.2 from G1 to G2/M phases and downregulates the expression of C-KIT mRNA and KIT protein in all three cell lines, particularly in HMC-1.1 and HMC-1.2 with activating mutations in KIT.

Summary: The proliferation of mast cells (MCs) is regulated by protein kinase C (PKC) redundancy, and inhibiting both PKC alpha and beta can prevent MC proliferation, providing new targets for the treatment of allergic diseases.
