The emerging roles of circRNAs in cancer and oncology

Lasse S. Kristensen et al.

Summary: Circular RNAs (circRNAs) are a novel class of primarily non-coding RNAs with increasingly recognized roles in cancer development and progression through diverse mechanisms of action. In this review, the authors summarize the current understanding of circRNA biogenesis, regulation, physiological functions and pathophysiological roles in cancer. They also discuss the clinical potential of circRNAs as biomarkers, therapeutic agents and drug targets in oncology, as well as the research controversies, technical issues, and biological knowledge gaps that need to be addressed.

circMRPS35 promotes malignant progression and cisplatin resistance in carcinoma

Peng Li et al.

Summary: This study identifies circMRPS35 as a highly expressed circRNA in HCC and shows that its knockdown inhibits cell proliferation, migration, invasion, and colony formation, as well as tumor growth. circMRPS35 regulates PTEN expression and degradation through miR-148a and STX3. Additionally, the circMRPS35-encoded peptide circMRPS35-168aa promotes cisplatin resistance in HCC.

Hsa_circ_0000098 is a novel therapeutic target that promotes hepatocellular carcinoma development and resistance to doxorubicin

Yi Li et al.

Summary: This study discovered that circ_0000098 is an oncogenic circRNA that promotes the development of HCC through the miR-383/MCUR1 axis. Currently, targeting circ_0000098 with DOX/sh-1@PLT holds promising and practical therapeutic potential for preventing DOX resistance in HCC.

Hepatocellular carcinoma

Josep M. Llovet et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global health challenge with rising incidence projected to exceed 1 million cases by 2025. Infection by hepatitis B and C viruses remains a major risk factor for HCC, while non-alcoholic steatohepatitis is emerging as a more common risk factor in Western countries. Advances in systemic therapies for HCC, including immunotherapies and targeted therapies, are expected to revolutionize the management of this disease.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Hsa_circ_0011385 knockdown represses cell proliferation in hepatocellular carcinoma

Chuangye Ni et al.

Summary: This study found that high expression of circ_0011385 in HCC is closely associated with tumor size, TNM stage, and prognosis, and its inhibition can prevent proliferation of HCC cells. Circ_0011385 acts as a sponge for miR-361-3p, regulating the mRNA expression of STC2, with its transcription being regulated by SP3. Therefore, circ_0011385 may serve as a new biomarker for HCC.

Summary: Recent findings have shown that RNA processing is systematically altered in cancer, with mutations in RNA splicing factor genes and the shortening of 3' untranslated regions widely observed. Additionally, evidence suggests that other types of RNAs, including circular RNAs, can contribute to tumorigenesis.
