A Two-Compartment Fermentation System to Quantify Strain-Specific Interactions in Microbial Co-Cultures

Summary: A two-compartment bioreactor system was developed to investigate microbial interactions in co-cultures. The system allowed for the exchange of amino acids and peptides between compartments. Synchronisation of glycolytic activity in yeast cells

Summary: Glycolysis is the central metabolic pathway in almost all cells and organisms, which can lead to oscillations at the level of individual cells, cell populations, or tissues. Coherent oscillations of metabolites are driven by glycolytic oscillations, particularly in biological systems like cardiomyocytes and pancreas. Studies on yeast have provided insight into the synchronization of glycolytic oscillations, which involve cell-to-cell communication through diffusion-mediated messenger molecules. Quorum sensing plays a role in synchronizing the collective oscillatory behavior of yeast cell populations, with individual cells continuing or ceasing to oscillate based on population density.

Summary: A mechanistic understanding of microorganisms in starter cultures is crucial for improving fermented milk products like yogurt. This study developed a synthetic medium for culturing microorganisms under defined conditions and used flow cytometry to measure their biomass. The study analyzed amino acid concentrations in the mono-cultures and co-cultures of L. bulgaricus and different strains of S. thermophilus, revealing that amino acid release rates in co-cultures were not equal to the sum of release rates in mono-cultures. Data-driven and pH-dependent models were developed to compare this release behavior. The study also identified specific amino acids, such as histidine, isoleucine, and arginine, that were affected by co-culture conditions. These findings contribute to the understanding of inter-species interactions and the targeted improvement of yogurt qualities.
