Current Insights into the Role of BRAF Inhibitors in Treatment of Melanoma

Understanding melanoma pathogenesis, BRAF conformations, and the problems with current FDA-approved inhibitors is crucial for new drug discovery and improving treatment effectiveness.

Resistance to Resveratrol Treatment in Experimental PTSD Is Associated with Abnormalities in Hepatic Metabolism of Glucocorticoids

Summary: This study found that post-traumatic stress disorder (PTSD) is associated with increased activity of hepatic 11 beta-HSD-1 and decreased activity of hepatic CYP3A. However, resveratrol, a natural polyphenol, has been shown to have protective effects against PTSD. Treatment with resveratrol in rats with PTSD resulted in two phenotypes, sensitive and resistant, with different effects on anxiety-like behavior and corticosterone concentration.

Hybrid Quinazoline 1,3,5-Triazines as Epidermal Growth Factor Receptor (EGFR) Inhibitors with Anticancer Activity: Design, Synthesis, and Computational Study

Summary: A series of hybrid quinazoline-1,3,5-triazine derivatives as EGFR inhibitors were synthesised and tested, showing activity against cancer cell lines with compounds 7c, 7d, 7e, and 7j being the most potent. In vitro and in silico techniques were used to elucidate essential substituent groups, with SAR studies performed for compound optimisation. Compound 7e showed promising activity against breast cancer in vivo and may serve as a lead compound for further anti-EGFR drug development.
