Molecular Characterization of TGF-Beta Gene Family in Buffalo to Identify Gene Duplication and Functional Mutations

Muhammad Saif-Ur Rehman et al.

Summary: The TGF-beta superfamily in buffalo was characterized through evolutionary, structural, and SNPs analyses to identify their functional effect. Multiple functions such as cell adhesion, motility, proliferation, apoptosis, and differentiation were found in the buffalo TGF-beta superfamily. The study also identified potential damaging mutations with functional consequences in buffalo growth, folliculogenesis, or embryogenesis.

Role of Peroxisome Proliferator-Activated Receptors (PPARs) in Energy Homeostasis of Dairy Animals: Exploiting Their Modulation through Nutrigenomic Interventions

Faiz-ul Hassan et al.

Summary: PPARs are nuclear receptors that regulate metabolic networks through nutrient-dependent transcriptional activation, but can be disrupted under metabolic stress. Nutrients such as polyunsaturated fatty acids, vitamins, and phytochemicals can modulate PPARs through various mechanisms, ultimately helping to prevent metabolic disorders.

FGF19 protects skeletal muscle against obesity-induced muscle atrophy, metabolic derangement and abnormal irisin levels via the AMPK/SIRT-1/PGC-α pathway

Ai Guo et al.

Summary: The study demonstrates that FGF19 administration can improve muscle loss and physical dysfunction in obesity and sarcopenic obesity (SO), as well as promote energy expenditure and restore metabolic derangement. Additionally, FGF19 exerts protective effects on skeletal muscle partly through the AMPK/SIRT-1/PGC-alpha signaling pathway, suggesting its potential as a therapeutic target for obesity and SO.

Comparative Genomic Characterization of Buffalo Fibronectin Type III Domain Proteins: Exploring the Novel Role of FNDC5/Irisin as a Ligand of Gonadal Receptors

Siwen Wu et al.

Summary: A total of 29 fibronectin type III domain (FN-III) protein genes were identified and characterized in the buffalo genome. Molecular and evolutionary analysis showed the well-conserved nature of FN-III proteins with diverse properties. The study also predicted the binding scores and interface residues of FNDC5/irisin as a ligand for various receptors, revealing its functional role in energy homeostasis, folliculogenesis, and spermatogenesis in buffalo.

Comparative Genomics, Evolutionary and Gene Regulatory Regions Analysis of Casein Gene Family in Bubalus bubalis

Saif Ur Rehman et al.

Summary: This study revealed the evolutionarily conserved nature of casein genes in buffalo, with greater amino acid sequence variation in river buffalo breeds that may affect milk trait regulation. Identified transcription activators and repressor sites in swamp buffalo suggest an association with lower expression of casein genes affecting milk production. Further functional studies are needed to evaluate the role of main motifs in controlling casein gene expression in buffalo.

FGF gene family characterization provides insights into its adaptive evolution in Carnivora

Qinguo Wei et al.

Summary: This study analyzed the FGF genes of 52 Carnivora species and found that the FGF gene family was divided into 10 subfamilies, subjected to purifying selection, and highly conserved in the evolutionary process of Carnivora. The phylogenetic comparative analyses indicated that habitat was one of the factors shaping the evolution of Carnivora FGF genes.

Hypoblast Formation in Bovine Embryos Does Not Depend on NANOG

Claudia Springer et al.

Summary: The role of NANOG in bovine embryo development was studied by knocking out the gene in fibroblast cells and producing NANOG knockout embryos via somatic cell nuclear transfer. The results showed that the FGF/MEK signaling pathway mediated by NANOG is not necessary for hypoblast formation in bovine embryos, suggesting the existence of another unknown pathway regulating hypoblast differentiation.

Genome-Wide Association Study Based on Random Regression Model Reveals Candidate Genes Associated with Longitudinal Data in Chinese Simmental Beef Cattle

Lili Du et al.

Summary: Traditional GWAS methods are limited to single-record traits, while the GWAS based on random regression model showed better performance in handling longitudinal traits. The study identified 37 significant SNPs and several candidate genes associated with body weight, which were enriched in signaling transduction pathway and lipid metabolism, suggesting potential molecular targets for genetic selection in cattle.
