Probing binding mode between sodium acid pyrophosphate and albumin: multi-spectroscopic and molecular docking analysis

Mechanistic and kinetic aspects of Natamycin interaction with serum albumin using spectroscopic and molecular docking methods

Summary: In this study, the interaction between natamycin (NT) and bovine serum albumin (BSA) was evaluated using surface plasmon resonance (SPR) and several spectroscopic techniques, accompanied by molecular docking analysis. The results showed that the fluorescence intensity of BSA decreased upon addition of NT, indicating complex formation through hybrid quenching. The binding constant between BSA and NT decreased with increasing temperature, and van der Waals forces and hydrogen bonds were found to be the main forces driving the interaction. Molecular docking analysis confirmed specific interaction sites between BSA and NT. The study also observed a decrease in binding constant between BSA and NT with increasing temperature.

Summary: This study systematically characterized the binding of ASK and BSA to understand the pharmaceutics properties of the ligand and the conformational changes of BSA upon the addition of ASK. The results indicated a temperature-dependent binding constant between ASK and BSA. Thermodynamic analysis revealed viable hydrophobic and hydrogen bonds in the moderate-affinity complex formation. CD spectroscopy showed changes in the secondary structure of BSA, with an increased content of alpha-helices upon the addition of ASK. AFM and DLS techniques demonstrated slight conformational shrinkage and the formation of large particles upon ASK binding. Molecular modeling suggested allosteric modulation of BSA structure in the vicinity of the tryptophan residue. Overall, the results of ASK-BSA interaction have significant implications for expanding the bio-applications of shikonin.
