Recent updates on 1,2,3-triazole-containing hybrids with in vivo therapeutic potential against cancers: A mini-review

Summary: The 1,2,3-triazole moiety, constructed by copper-catalyzed azide-alkyne cycloaddition, serves as a versatile linker and a useful pharmacophore with diverse biological properties. It easily interacts with enzymes and receptors in cancer cells, inhibiting cell proliferation, arresting cell cycle, and inducing apoptosis. 1,2,3-triazole-containing hybrids can exert dual or multiple anticancer mechanisms, making them valuable in the development of novel anticancer agents. This review summarizes the in vivo anticancer efficacy and mechanisms of action of 1,2,3-triazole-containing hybrids, providing a roadmap for further exploration of effective candidates.

Click chemistry inspired syntheses of new amide linked 1,2,3-triazoles from naphthols: biological evaluation and in silico computational study

Summary: A small library of 1,2,3-triazole derivatives derived from naphthols has been efficiently synthesized through the click chemistry approach for the search of new active molecules. The synthesized compounds were evaluated for their antifungal, antioxidant, and antitubercular activities. Furthermore, a molecular docking study was conducted to understand the binding affinity and interactions between the enzyme and the synthesized derivatives, revealing a significant correlation between the binding score and biological activity of these compounds. The results suggest that the 1,2,3-triazole derivatives may serve as potential candidates for the development of novel therapeutic agents.

Substituted naphthoxy-phthalonitrile derivatives: Synthesis, substituent effects, DFT, TD-DFT Calculations, antimicrobial properties and DNA interaction studies

Summary: In this study, substituted-naphthol derivatives were synthesized and reacted with phthalonitrile derivatives to obtain the desired compounds. The structures of the synthesized compounds were characterized using spectroscopic techniques. The synthesized compounds showed strong antimicrobial activity against Gram-positive bacteria and exhibited some inhibitory effects on Gram-negative bacteria and yeast. The compounds also caused changes in the structure and mobility of plasmid DNA. Theoretical calculations were performed to analyze the structures and properties of the compounds.

Preparation of antioxidant-biodegradable poly(acrylic acid-co-2-hydroxyethyl methacrylate) hydrogel using rutin as a crosslinker: Drug release and anticancer activity

Summary: This study developed a drug carrier, poly(acrylic acid-co-2-hydroxyethyl methacrylate)/rutin (RTN hydrogel), using the natural crosslinker rutin. The RTN hydrogel exhibited in vitro biodegradability and antioxidant properties. It adsorbed a maximum of 32.96±1.83 mg/g doxorubicin (DOX) and released the drug in normal and cancer cell media. The effect on triple negative breast cancer (TNBC) cell lines was also investigated, showing promising potential as a high-impact carrier in drug distribution systems.

Synthesis, antimicrobial evaluation and docking studies of fluorinated imine linked 1,2,3-triazoles

Summary: A diverse series of imine linked 1,2,3-triazole hybrids were synthesized via Cu(I)-promoted click reaction, and their structures were characterized using various spectral techniques. Among them, 7d exhibited the highest antimicrobial activity against R. oryzae and S. aureus. Docking studies revealed that some triazole hybrids showed high potency towards E. coli DNA gyrase and C. albicans sterol 14-alpha demethylase.

A 1,2,3-Triazole Derivative of Quinazoline Exhibits Antitumor Activity by Tethering RNF168 to SQSTM1/P62

Summary: We synthesized a series of 1,2,3-triazole derivatives of quinazoline and found that compound 5a exhibited the strongest cytotoxicity against HCT-116 cells. Mechanistic studies revealed that 5a promotes the interaction between P62 and RNF168, resulting in decreased tolerance of HCT-116 cells to X-ray radiation and suppression of tumor growth in mouse xenograft models.

Summary: In this study, twelve novel anti-tuberculosis compounds were efficiently synthesized via click chemistry and their activities against tuberculosis, antioxidant, cytotoxicity, and enzyme were evaluated. The results showed that these compounds exhibited good activity against tuberculosis, moderate to low antioxidant activity, and good to moderate cytotoxicity against lung cancer cell lines. Molecular docking studies also revealed their interaction with DprE1 protein of Mycobacterium tuberculosis.
