In-silico structural studies on anti-inflammatory activity of phytocompounds from the genus Andrographis

Structure and dynamics of 5-lipoxygenase and its complexes - a molecular dynamics simulation study

Summary: Molecular dynamics simulations were used to investigate the structure and dynamics of human 5-LOX with and without ligands. The simulations revealed changes in the protein's dynamical properties and other structural features upon ligand binding. Furthermore, the study demonstrated the significant central bending in the protein and ruled out the suspected water coordination to the metal ion. Additionally, the binding free energy of Zileuton and CAPE1 inhibitors to 5-LOX was estimated, revealing a stronger binding potential for CAPE1 compared to Zileuton, consistent with their respective IC50 values.

NSAIDs and Cancer Resolution: New Paradigms beyond Cyclooxygenase

Summary: Acute inflammation is an adaptive host defense mechanism, but unresolved or chronic inflammation can lead to various maladies, including cancer. Nonsteroidal anti-inflammatory drugs (NSAIDs) such as aspirin have been shown to reduce the risk and mortality of many cancers. Besides targeting cyclooxygenase-2 (COX-2), NSAIDs also affect other important targets, and specialized pro-resolving mediators (SPMs) promote the resolution of inflammation and tissue healing, showing potential in inhibiting cancer growth and metastasis.
