Weight Loss and Alzheimer's Disease in Down Syndrome

Victoria Fleming et al.

Summary: This study aimed to determine if unintentional weight loss is part of Alzheimer's disease (AD) in adults with Down syndrome (DS). The results showed that unintentional weight loss is associated with A beta deposition and occurs prior to the onset of AD dementia in DS, indicating that it may be a useful sign of AD in this population.

Exploring semantic verbal fluency patterns and their relationship to age and Alzheimer's disease in adults with Down syndrome

Farah Mgaieth et al.

Summary: Adults with Down syndrome have a high risk of developing Alzheimer's disease, showing poor verbal fluency and semantic memory similar to the general population in the preclinical phase. This study investigated the semantic fluency performance in adults with DS and its relationship to age, AD, and blood biomarkers. The results suggest that semantic fluency may serve as an early indicator of cognitive decline and be associated with biomarkers in adults with DS.

Basal forebrain atrophy along the Alzheimer's disease continuum in adults with Down syndrome

Mateus Rozalem Aranha et al.

Summary: This study reveals that basal forebrain (BF) degeneration is an important biomarker of Alzheimer's disease (AD) in individuals with Down syndrome (DS), and it is associated with cognitive decline, changes in cerebrospinal fluid and plasma biomarkers, and hippocampal volume reduction.

Cross-sectional versus longitudinal cognitive assessments for the diagnosis of symptomatic Alzheimer's disease in adults with Down syndrome

Laura Videla et al.

Summary: This study compares the performance of baseline and longitudinal neuropsychological assessments for the diagnosis of symptomatic Alzheimer's disease (AD) in individuals with Down syndrome (DS). The results show that baseline assessments have better diagnostic performance for AD dementia and prodromal AD than the 1-year intra-individual cognitive decline. Longer follow-up periods improve the diagnostic performance of intra-individual cognitive decline.

Association of biological sex with clinical outcomes and biomarkers of Alzheimer's disease in adults with Down syndrome

M. Florencia Iulita et al.

Summary: The study highlights the importance of conducting sex-specific research in Alzheimer's disease, particularly in individuals with Down syndrome who have a high genetic risk. The findings suggest that while there are similarities in prevalence and cognitive outcomes between males and females, males may experience lower recall test scores. Additionally, the presence of the apolipoprotein.4 allele may affect the age at diagnosis in females.

Selective Impairment of Long-Range Default Mode Network Functional Connectivity as a Biomarker for Preclinical Alzheimer's Disease in People with Down Syndrome

Natalie D. DiProspero et al.

Summary: Lower long-range functional connectivity within the default mode network predicts Alzheimer's disease diagnosis and cognitive decline in individuals with Down syndrome. Amyloid-beta accumulation in the inferior parietal cortex is associated with reduced regional DMN functional connectivity, indicating a potential biomarker for Alzheimer's disease in this population.

Proposed diagnostic criteria for mild cognitive impairment in Down syndrome population

Susanna Esteba-Castillo et al.

Summary: The study aimed to describe cognitive and neuropsychological patterns associated with MCI in individuals with Down syndrome. The results indicated that BRI and delayed verbal memory were the best predictors of group membership. The diagnosis of MCI in this population should consider changes in BRI and abstract thinking ability, along with patterns in delayed verbal memory.

Summary: This study used diffusion-weighted imaging and connectomic modeling to assess the predictive ability of brain amyloid plaque burden, baseline cognition, and longitudinal cognitive change in individuals with Down's syndrome. The findings suggest that graph theory metrics based on the structural connectome are effective predictors of global amyloid deposition, and the connection density of the structural network at baseline is a promising predictor of current cognitive performance.

Amyloid-beta and tau deposition influences cognitive and functional decline in Down syndrome

Monika Grigorova et al.

Summary: This study examines the independent effects of tau and amyloid-beta on cognitive and functional performance changes, as well as their synergistic relationship in exacerbating decline in aging Down syndrome (DS) individuals. The findings reveal that baseline tau deposition is a significant predictor of cognitive and functional decline, above and beyond the impact of amyloid-beta status.

Neural correlates of episodic memory in adults with Down syndrome and Alzheimer's disease

Bessy Benejam et al.

Summary: This study aimed to investigate the neuroanatomical correlates of episodic memory in adults with Down syndrome and symptomatic Alzheimer's disease. The results showed that different types of episodic memory were associated with cortical atrophy in specific brain regions. These findings further support the similarities between Down syndrome-associated Alzheimer's disease and that in the general population.

Physical activity, memory function, and hippocampal volume in adults with Down syndrome

Role of tau deposition in early cognitive decline in Down syndrome

Sigan L. Hartley et al.

Summary: High tau levels may indicate early prodromal Alzheimer's disease in Down syndrome, similar to late-onset and autosomal dominant Alzheimer's disease. The study found that in individuals with A beta positivity, tau SUVR was negatively associated with episodic memory performance. Individuals with both A beta positivity and high tau levels performed significantly worse in episodic memory compared to those with low tau levels.

Hypersynchronized Magnetoencephalography Brain Networks in Patients with Mild Cognitive Impairment and Alzheimer's Disease in Down Syndrome

Summary: The study findings of the AD-continuum in individuals with Down syndrome (DS) could assist in characterizing disease stages and support the role of excitatory/inhibitory imbalance in Alzheimer's disease (AD). The pattern of functional connectivity (FC) hypersynchronization in DS individuals resembles the reported AD FC evolution in typically developing populations, and the conclusions of this study could aid in predicting and characterizing DS individuals with a higher risk of developing dementia.

Cognitive and Behavioral Domains That Reliably Differentiate Normal Aging and Dementia in Down Syndrome

Jordan P. Harp et al.

Summary: This study employed principal components analysis (PCA) and classification and regression tree (CART) analysis to identify an abbreviated dementia classification battery for individuals with Down syndrome in primary care settings. Results showed that the sum of cognitive scores for DLD and community subdomain scores for Vineland-II were the best classifiers for dementia diagnosis. The sensitivity of PCA was 80% and specificity was 70%, while CART had a sensitivity of 100% and specificity of 81%. The findings support the use of an abbreviated dementia screening battery for at-risk individuals with Down syndrome.
