Ecological Niche-Inspired Genome Mining Leads to the Discovery of Crop-Protecting Nonribosomal Lipopeptides Featuring a Transient Amino Acid Building Block

Summary: Investigating the ecological context of microbial predator-prey interactions helps in identifying microorganisms that produce secondary metabolites to evade predation or kill the predator. By using genomics-based methods, new biosynthetic gene clusters can be discovered for the development of antimicrobials. This study describes the identification of keanumycins, which enable bacteria to evade predation and exhibit antimycotic activity against plant pathogens.

Deciphering chemical logic of fungal natural product biosynthesis through heterologous expression and genome mining

Summary: This review highlights key examples in the integration of heterologous expression into genome mining and biosynthetic investigations in the last decade. By choosing appropriate heterologous hosts, prioritizing BGCs for structural novelty, and using shunt products from heterologous expression, important insights into the chemical logic of biosynthesis can be revealed. This study demonstrates the usefulness and limitations of heterologous biosynthesis in fungal natural product discovery.

Genome mining for unknown-unknown natural products

Summary: Genome mining revealed a conserved fungal biosynthetic pathway that lacked a core enzyme. Heterologous expression identified a highly modified cyclo-arginine-tyrosine dipeptide as the associated natural product. Biochemical characterization led to the discovery of a new arginine-containing cyclodipeptide synthase (RCDPS) that showed no sequence homology to known enzymes and is widely encoded in fungal genomes. Further research on the biosynthetic pathway led to the discovery of new compounds whose structures would not have been predicted without the knowledge of RCDPS function.

Induced secondary metabolites of the beneficial fungus Trichoderma harzianum M10 through OSMAC approach

Summary: A bioactive chromone derivative with antibiotic and antitumor activities was isolated from Trichoderma harzianum M10 using OSMAC strategy. It showed significant growth inhibition against Rhizoctonia solani and reduced the viability of colorectal cancer cells in a concentration-dependent manner. Metabolomic analysis also revealed the modulation of several compounds by the cultivating conditions.

Discovery and characterization of a terpene biosynthetic pathway featuring a norbornene-forming Diels-Alderase

Summary: This study reports the discovery of a norbornene synthase SdnG and its role in the biosynthesis of the antifungal natural product sordarin. The reconstitution of sordarin biosynthesis reveals a concise oxidative strategy used by Nature to transform a hydrocarbon precursor into a highly functionalized substrate for SdnG. It was found that SdnG generates the norbornene core of sordarin and accelerates the intramolecular Diels-Alder reaction.

Article Unusual (2R,6R)-bicyclo[3.1.1]heptane ring construction in fungal α-trans-bergamotene biosynthesis

Summary: This study isolated and characterized various bergamotene derivatives from a marine fungus, and identified their corresponding synthases. The proposed cyclization mechanism of bergamotene was supported by molecular docking, and the biocontrol potential of these compounds was demonstrated.

Copper starvation induces antimicrobial isocyanide integrated into two distinct biosynthetic pathways in fungi

Summary: The genomes of filamentous fungi include many biosynthetic gene clusters of unknown function. In this study, the authors show that copper starvation induces expression of an enzyme (CrmA) that generates a valine-derived isocyanide participating in two different pathways for biosynthesis of acylated sugar alcohols and modified ergot alkaloids.

Naphthalene derivatives and halogenate quinoline from the coral-derived fungus Trichoderma harzianum (XS-20090075) through OSMAC approach

Summary: The OSMAC approach was used to study the changes in secondary metabolites of the soft coral-derived fungus Trichoderma harzianum by using two different cultures. Three new compounds were isolated and identified, with the structure of one compound confirmed by single-crystal X-ray diffraction. This study also reported the first halogenate quinoline derivative isolated from the genus of Trichoderma.

The unique aziridine ring opening by a HAD-type hydrolase and oxidative decarboxylation by a non-heme iron oxygenase are key steps in this process.

AoiQ, a flavin-dependent halogenase, catalyzes gem-dichlorination of 1,3-diketone substrates in the biosynthesis of dichlorodiaporthin, representing the first biochemically reconstituted FDH that can halogenate an enolizable sp(3)-hybridized carbon atom.

Harzianic Acid from Trichoderma afroharzianum Is a Natural Product Inhibitor of Acetohydroxyacid Synthase

Summary: Acetohydroxyacid synthase (AHAS) is a crucial enzyme in the branched-chain amino acid biosynthetic pathway and harzianic acid (HA) produced by the biocontrol fungus has been identified as a natural product inhibitor of AHAS. HA selectively inhibits fungal AHAS, including resistant mutants, through a competitive inhibition mechanism different from known commercial inhibitors. The biosynthesis of HA by the fungus in the rhizosphere is proposed as an important role based on the study data.

New strategies for discovering secondary metabolites of Trichoderma in recent years are also discussed.

Genome mining of cryptic tetronate natural products from a PKS-NRPS encoding gene cluster in Trichoderma harzianum t-22

Summary: In this study, a PKS-NRPS encoding gene cluster from Trichoderma harzianum was heterologously reconstituted in Aspergillus nidulans, leading to the isolation of six new tetronate natural products. New tetronate compounds trihazone A-F were characterized using HRESIMS and NMR data, with three of them containing an exocyclic olefin derived from ThnC-dependent dioxygenase.
