Structural Basis for Chaperone-Independent Ubiquitination of Tau Protein by Its E3 Ligase CHIP

Francesca Munari et al.

Summary: The study reveals that E3 ligase CHIP can ubiquitinate Tau protein at multiple sites, even in the absence of molecular chaperones, providing mechanistic insight into the chaperone-independent engagement of a disordered protein by its E3 ligase.

Tau liquid-liquid phase separation in neurodegenerative diseases

Solomiia Boyko et al.

Summary: The aggregation of tau protein is important in neurodegenerative diseases, including Alzheimer's disease. Recent studies have found that tau, similar to other proteins associated with neurodegeneration, has a high propensity for liquid-liquid phase separation (LLPS). This article discusses the evidence for tau LLPS in vitro, the molecular mechanisms involved, and the effects of post-translational modifications and pathogenic mutations on tau phase separation. The article also explores recent studies on tau LLPS in cells, providing insights into the relationship between LLPS and neurodegeneration in tauopathies.

Ubiquitination of Alzheimer's-related tau protein affects liquid-liquid phase separation in a site- and cofactor-dependent manner

Francesca Parolini et al.

Summary: The formation of biomolecular condensates plays a crucial role in neuronal physiology and neurodegeneration. This study reveals that ubiquitination can regulate the behavior of tau protein during condensate formation and potentially influence the physiological and pathological states of tau in cellular condensates.

Neuroprotective Effects of Coffee Bioactive Compounds: A Review

Katarzyna Socala et al.

Summary: Coffee contains various bioactive components that are beneficial for human health, such as caffeine, polyphenols, and alkaloids. Research suggests that regular coffee consumption can reduce the risk of cardiovascular diseases, type 2 diabetes, obesity, and certain types of cancer, while also potentially lowering the risk of neurodegenerative conditions like Alzheimer's and Parkinson's diseases. Further studies are needed to fully understand the mechanisms behind these effects and the therapeutic applications of coffee bioactive components.

Carlotta Ciaramelli et al.

Summary: In this study, the metabolic profile of Lavado cocoa was characterized using NMR spectroscopy for the first time, revealing its anti-amyloidogenic activity. The results showed that catechins and theobromine in Lavado cocoa were key chemical components responsible for inhibiting Aβ peptide aggregation and toxicity in human neuroblastoma cells.

HT-SuMD: making molecular dynamics simulations suitable for fragment-based screening. A comparative study with NMR

Francesca Ferrari et al.

Summary: Fragment-based lead discovery is an efficient method for developing new drugs. The new computational protocol HT-SuMD allows for automatic screening of thousands of fragments, showing remarkable agreement with NMR-based screening. Virtual screening on a larger library of fragments confirmed all selected hits as Bcl-X-L binders, representing the largest computational fragment screening based on MD to date.
