Engineering of the Substrate Pocket of α-ketoglutaric Semialdehyde Dehydrogenase for Improving the Activity toward 3-hydroxypropanal

Summary: An engineered variant of aldehyde dehydrogenase, 2C10, was obtained from Azospirillum brasilense, which showed a 4.65-fold increase in catalytic activity towards 3-HPA compared to the wild-type enzyme. Culturing Pseudomonas denitrificans with 2C10 resulted in approximately 30% higher 3-HP production compared to wild-type KGSADH. Molecular dynamics simulations suggested that 2C10 has a less flexible and smaller binding pocket for aldehyde substrates compared to the wild-type enzyme.

Deciphering glutamate and aspartate metabolism to improve production of succinate in Escherichia coli

Engineering the Cad pathway in Escherichia coli to produce glutarate from l-lysine

Production of Succinic Acid from Amino Acids in Escherichia coli

By inactivating succinate dehydrogenase and conducting metabolic evolution, a mutant strain capable of efficiently metabolizing aspartate was developed. After further optimization through evolving, a resulting strain was able to produce succinic acid with high yield and productivity. This study presents a promising avenue for a biorefinery platform based on renewable amino acids.

Overcoming glutamate auxotrophy in Escherichia coli itaconate overproducer by the Weimberg pathway

Summary: Engineering efficient itaconate producers often requires elimination of isocitrate dehydrogenase to preserve cis-aconitate, leading to auxotrophy for 2-ketoglutarate. This study presents a novel approach by introducing the Weimberg pathway for 2-ketoglutarate biosynthesis, overcoming the need for glutamate supplementation. Through careful tuning of xylose concentration, the final strain produced 20 g/L of itaconate in minimal medium without the need for antibiotics.
