Genetic variations in GABA metabolism and epilepsy

Yan Feng et al.

Summary: Epilepsy is a brain disorder caused by an imbalance between neuronal excitation and inhibition. The most important inhibitory neurotransmitter in the brain, gamma-aminobutyric acid (GABA), plays a crucial role in the occurrence and development of epilepsy. Abnormalities in various aspects of GABA metabolism, including synthesis, transport, genes encoding GABA receptors, and inactivation, can contribute to epilepsy. Genetic mutations of GABA receptor encoding genes, such as GABRA1, GABRA2, GABRA5, GABRB1, GABRB2, GABRB3, GABRG2, and GABBR2, are commonly associated with epilepsy, leading to different clinical phenotypes by reducing receptor expression and GABA-evoked potentials. Mutations in the ABAT and ALDH5A1 genes, which encode enzymes involved in GABA metabolism, can result in symptoms related to their deficiency, such as epilepsy and cognitive impairment. Most genetic variations in GABA metabolism are accompanied by developmental disorders. Understanding the relationship between genetic variations in GABA metabolism and epilepsy is important for accurate diagnosis and treatment.

Endothelial Dysfunction in Atherosclerotic Cardiovascular Diseases and Beyond: From Mechanism to Pharmacotherapies

Suowen Xu et al.

Summary: The endothelium, a cellular monolayer lining the blood vessel wall, plays a critical role in maintaining multiorgan health and homeostasis. Dysfunction of the vascular endothelium is associated with various diseases and is implicated in severe coronavirus disease 2019. Therapeutic approaches targeting endothelial dysfunction may have potential for treatment of cardiovascular and many other diseases.

Anticonvulsant effect of pterostilbene and its influence on the anxiety- and depression-like behavior in the pentetrazol-kindled mice: behavioral, biochemical, and molecular studies

Dorota Nieoczym et al.

Summary: Pterostilbene exhibits anticonvulsant effects in a PTZ-induced kindling mouse model, possibly mediated through increasing GABA concentrations and reducing mRNA expression of BDNF and TNF-alpha. It shows potential as a candidate for a new antiseizure drug.

Immune Cells in the BBB Disruption After Acute Ischemic Stroke: Targets for Immune Therapy?

Yan-mei Qiu et al.

Summary: Blood-Brain Barrier (BBB) disruption is a crucial pathophysiological process in acute ischemic stroke, where the rapid activation of immune cells plays a critical role. Infiltrating immune cells increase BBB permeability, causing microvascular disorder, but also promote BBB repair and angiogenesis. Understanding the inflammatory mechanisms behind BBB disruption can lead to the development of more effective therapeutics for acute ischemic stroke.

Effects of Antidepressant Treatment on Neurotrophic Factors (BDNF and IGF-1) in Patients with Major Depressive Disorder (MDD)

Anna Mosiolek et al.

Summary: Major depressive disorder (MDD) is a multifactorial disease with ongoing research on the role of neurotrophic factors in its pathophysiology. Antidepressant treatment can modify serum levels of neurotrophins, such as BDNF, leading to clinical improvement of MDD. Further research is needed to understand the correlation between different antidepressant drugs and neurotrophic factor serum levels.

Activation of Nrf2/HO-1 signaling: An important molecular mechanism of herbal medicine in the treatment of atherosclerosis via the protection of vascular endothelial cells from oxidative stress

Qing Zhang et al.

Summary: This study summarized natural compounds that exert protective effects against oxidative stress in vascular endothelial cells (ECs), including phenylpropanoids, flavonoids, terpenoids, and alkaloids. These compounds alleviate EC apoptosis by activating the Nrf2/HO-1 signaling pathway, showing potential for the treatment of atherosclerosis.

Linking Oxidative Stress and Proteinopathy in Alzheimer's Disease

Chanchal Sharma et al.

Summary: Proteinopathy and excessive production of reactive oxygen species are key factors contributing to neuronal toxicity in Alzheimer's disease. Beta-amyloid and tau proteins play a significant role in proteinopathy in AD, which is closely related to ROS production. The mechanisms of proteinopathy and ROS production work together to drive the pathology observed in aging brains, ultimately leading to oxidative stress and neurodegeneration.

NADPH Oxidases (NOX): An Overview from Discovery, Molecular Mechanisms to Physiology and Pathology

Annelise Vermot et al.

Summary: NADPH oxidase (NOX) enzymes, which produce reactive oxygen species (ROS), play crucial roles in physiological processes such as immune defense, protein post-translational processing, cellular signaling, gene expression regulation, and cell differentiation. However, dysregulation of NOX can lead to various severe pathologies, making them potential therapeutic targets. Understanding the molecular mechanisms behind NOX regulation and ROS production is essential for investigating human diseases and developing new treatments.

Oxidative Stress and Beta Amyloid in Alzheimer's Disease. Which Comes First: The Chicken or the Egg?

Elena Tamagno et al.

Summary: The pathogenesis of Alzheimer's disease involves the accumulation of beta amyloid and vulnerability of the brain to oxidative stress, which are linked to each other. It is difficult to determine which comes first, Aβ or oxidative stress. Evidence suggests that oxidative stress occurs early in the development of Alzheimer's disease and plays a crucial role in the manifestation of clinical and pathological symptoms.

Summary: Pattern recognition receptors (PRRs) are a class of receptors that can directly recognize specific molecular structures on pathogens, apoptotic host cells, and damaged senescent cells, bridging nonspecific immunity and specific immunity. Through the recognition and binding of ligands, PRRs can produce immunoprotective effects against infections and tumors. Research on PRRs and their ligands has greatly enhanced understanding of the signaling pathways involved, providing insights for the treatment of immune-related diseases and tumors.
