Design, Synthesis, and Fungicidal Evaluation of Novel N-Methoxy Pyrazole-4-Carboxamides as Potent Succinate Dehydrogenase Inhibitors

Summary: Based on the binding mode of pydiflumetofen with SDH, the carbon-carbon double bond was introduced into pydiflumetofen to produce target compounds 6a-6o. The SAR study showed that the 2-position and 4-position in the terminal benzene were positively correlated with increased activity. Compound 6o exhibited good control effects against WPM, CPM, and SCR, showing its broad-spectrum property. Especially, compound 6o showed higher control effects against CPM and SCR compared to pydiflumetofen, indicating its potential for further development.

Multiple Mutations in SDHB and SDHC2 Subunits Confer Resistance to the Succinate Dehydrogenase Inhibitor Cyclobutrifluram in Fusarium fujikuroi

Summary: Fusarium fujikuroi, a dominant phytopathogenic fungus causing rice bakanae disease, is shown to be inhibited by the novel succinate dehydrogenase inhibitor cyclobutrifluram. Mutations in FfSdhB (H248L/Y) and FfSdhC2 (G80R or A83V) were found to confer resistance to cyclobutrifluram in F. fujikuroi, leading to decreased affinity between the drug and FfSdhs.

Molecular Basis for the Selectivity of the Succinate Dehydrogenase Inhibitor Cyflumetofen between Pest and Predatory Mites

Summary: Acaricides that inhibit mitochondrial succinate dehydrogenase (SDHI) have strong control over phytophagous mites but limited toxicity to predatory mites. This study reveals that the selectivity of SDHI acaricides is mainly due to the stronger inhibition on the SDH target in spider mites compared to predatory mites. The findings provide insights for the rational design of selective acaricides in integrated pest management.

Antifungal Drug Resistance: An Emergent Health Threat

Summary: Fungal infections, especially drug-resistant forms like Candida and Aspergillus spp., are increasing in prevalence and causing severe diseases. This is primarily due to the growing number of immunocompromised patients and the emergence of resistant strains. Design, synthesis, and fungicidal activity of novel N-substituted piperazine-containing phenylpyridines against cucumber downy mildew

Summary: This study presents the design and synthesis of a series of novel phenylpyridine derivatives. Bioassays demonstrated that most of these compounds exhibited excellent fungicidal activities against cucumber downy mildew (CDM). Among the phenylpyridine compounds, 2-(4-(4-(tert-butyl) benzyl)piperazin-1-yl)-6-phenylnicotinonitrile (C8) showed the highest activity, surpassing that of commercial fungicides. The molecular electrostatic potential analysis of compound C8 suggested its potential for hydrogen bonding and electrostatic interactions.
