Drug repurposing: a systematic review on root causes, barriers and facilitators

Nithya Krishnamurthy et al.

Summary: This article systematically reviews the root causes, facilitators, and barriers of drug repurposing. The study finds that promising drugs are often shelved due to insufficient efficacy, poor market prospects, and industry consolidation. Inadequate resources, data access, and IP negotiation challenges are key barriers to the full potential of drug repurposing. Multi-partner collaborations, access to compound databases, and tax incentives are key facilitators for repurposing.

Yajie Meng et al.

Summary: The study focuses on developing a Human Drug Virus Database (HDVD) by collecting drug-virus interaction entries and proposing a new approach called SCPMF to identify new drug-virus interactions for drug repurposing. SCPMF outperforms several recent approaches with reliable prediction performance, as demonstrated on two different datasets. Furthermore, SCPMF-predicted drug candidates of COVID-19 confirm the accuracy and reliability of the method.
