Multifunctional low temperature-cured PVA/PVP/citric acid-based hydrogel forming microarray patches: Physicochemical characteristics and hydrophilic drug interaction

Summary: The characteristics of multifunctional polymeric hydrogel-forming microarray patches based on poly(vinyl alcohol)/poly(vinylpyrrolidone)/citric acid composite crosslinked at 80 degrees C were investigated. The swelling study showed that this composite possesses a higher swelling degree than the same polymer heated at 130 degrees C due to a lower crosslink density, which was then confirmed by FTIR examination. The patch can function both as a means to sample model hydrophilic drugs from the skin and to deliver them when combined with a melt-type polyethylene glycol reservoir.

Ring inserts as a useful strategy to prepare tip-loaded microneedles for long-acting drug delivery with application in HIV pre-exposure prophylaxis

Summary: Microneedle array patches (MAPs), including dissolving MAPs, play an increasingly important role in transdermal drug delivery for the management of multiple diseases. However, challenges exist in the manufacturing process, such as the presence of bubbles and lack of uniformity in microneedle formation. In this study, a simple method using ring inserts was developed to produce tip-loaded MAPs. The obtained MAPs showed high uniformity in microneedle formation and suitable insertion capability. Pharmacokinetic studies in rats demonstrated that these MAPs could maintain therapeutic plasma levels of the drug for 14 days.

Summary: This article introduces the application of hydrogel-forming microarray patches (HF-MAPs) for minimally invasive, pain-free and prolonged drug delivery. The use of hydroxypropyl-I3-cyclodextrin (HP-I3-CD) to enhance the solubility of cabotegravir sodium (CAB-Na) is studied for the first time, and its effect on intradermal delivery via HF-MAPs is investigated. The results show that the HF-MAPs can effectively deliver CAB-Na drugs both ex vivo and in vivo.
