Direct cyclodextrin based powder extrusion 3D printing of budesonide loaded mini-tablets for the treatment of eosinophilic colitis in paediatric patients

Monica Pistone et al.

Summary: The study aimed to create a solid oral formulation of budesonide (BD) loaded with direct powder extrusion (DPE) 3D printing and fluid bed coating techniques for the treatment of eosinophilic colitis (EC) in paediatric patients. BD, the preferred medication for EC treatment, has poor water solubility and low absorption. The DPE 3D approach was used to create Mini-Tablets (MTs) suitable for paediatric patients, and the inclusion of hydroxypropyl-beta-cyclodextrin improved the solubility and dissolution rate of BD. The MTs were coated with an enteric polymer to achieve site-specific drug release at the intestinal level.

Direct cyclodextrin-based powder extrusion 3D printing for one-step production of the BCS class II model drug niclosamide

Monica Pistone et al.

Summary: This study successfully printed tablets containing Niclosamide using the direct powder extrusion 3D printing technique. This innovative method overcomes the limitations of previous techniques and demonstrates the potential to produce personalized pharmaceutical forms directly from powders. The tablets showed good characteristics in terms of solubility, drug release, stability, and mechanical properties.

Summary: This study examined the potency and delivery of clobetasol-17-propionate from an electrospun mucoadhesive patch using cell monolayers and oral mucosal equivalents. OMEs were found to be more suitable than cell monolayers for evaluating toxicity and drug delivery, and the patch-delivered clobetasol-17-propionate showed efficacy in treating OLP.

Spray-dried mucoadhesive microparticles based on S-protected thiolated hydroxypropyl-β-cyclodextrin for budesonide nasal delivery

Giuseppe Francesco Racaniello et al.

Summary: This study demonstrated that thiolated hydroxypropyl-beta-cyclodextrin can improve the water solubility of Budesonide and be used to prepare mucoadhesive microparticles for intranasal delivery. The spray-dried microparticles showed good stability, rapid dissolution, and prolonged mucosal residence time, making them a promising tool for the nasal delivery of poorly water-soluble corticosteroids.
