Pharmacokinetics of Orally Applied Cannabinoids and Medical Marijuana Extracts in Mouse Nervous Tissue and Plasma: Relevance for Pain Treatment

Cristiana Dumbraveanu et al.

Summary: Cannabis plants contain various bioactive substances, and THC and CBD are the most extensively studied. It remains unclear how the lesser investigated compounds in plant extracts affect THC or CBD's bioavailability or biological effects. A pilot study was conducted to compare THC concentrations in plasma, spinal cord, and brain after oral administration of THC with medical marijuana extracts rich in THC or depleted of THC. The study found that THC levels were higher in mice receiving the THC-rich extract. Surprisingly, only CBD, not THC, alleviated mechanical hypersensitivity in a mouse nerve injury model, suggesting CBD as a potential analgesic compound with fewer unwanted psychoactive effects.

A naturalistic study of orally administered vs. inhaled legal market cannabis: cannabinoids exposure, intoxication, and impairment

L. Cinnamon Bidwell et al.

Summary: This study aimed to characterize naturalistic use of legal market flower and edible products by examining associations among blood cannabinoids and amount of THC consumed as well as physiological, cognitive, and subjective effects in users of edible and flower forms. The findings suggest few differences in intoxication and impairment between edible and flower cannabis users after ad libitum use.

Cannabinoid tetrad effects of oral Δ9-tetrahydrocannabinol (THC) and cannabidiol (CBD) in male and female rats: sex, dose-effects and time course evaluations

Catherine F. Moore et al.

Summary: The study examined the effects of orally administered THC and CBD on pain sensitivity, body temperature, locomotor activity, and catalepsy in rats. THC produced long lasting effects of antinociception, hypothermia, hyper- and hypolocomotion, and catalepsy, while CBD had modest effects on pain sensitivity and sex-dependent effects on body temperature and locomotor activity.

Prevalence and use of cannabis products and routes of administration among youth and young adults in Canada and the United States: A systematic review

Elle Wadsworth et al.

Summary: This systematic review summarizes the prevalence of routes of administration and cannabis products used among youth and young adults in Canada and the United States. The review found that smoking had the highest prevalence rates among youth and young adults, but rates of use appeared to reduce over time, while vaping appeared to increase over time. Fewer studies focused on oral or dabbed cannabis, but those that did reported prevalence estimates of approximately a third among recent cannabis consumers. The findings highlight the need for guidance and education on different types of cannabis products and routes of administration for youth.

Sex differences in the acute effects of oral and vaporized cannabis among healthy adults

Dennis J. Sholler et al.

Summary: Research comparing the acute effects of oral and vaporized cannabis in occasional users found that females exhibited stronger drug effects in some aspects, especially when controlling for body weight. Recommendations include starting with lower doses for females and being aware of potential acute anxiogenic reactions when using cannabinoids.

Sex, species and age: Effects of rodent demographics on the pharmacology of Δ9-tetrahydrocanabinol

Jenny L. Wiley et al.

Summary: The study investigated the effects of age, sex, and rodent species on the psychoactive impact of intraperitoneal THC and its metabolite 11-OH-THC. The results show that 11-OH-THC exhibited higher potency in some aspects compared to THC, suggesting that species is a crucial design consideration in preclinical studies of phytocannabinoids.

Pharmacokinetics and central accumulation of delta-9-tetrahydrocannabinol (THC) and its bioactive metabolites are influenced by route of administration and sex in rats

Samantha L. Baglot et al.

Summary: Up to a third of North Americans report using cannabis in the prior month, most commonly through inhalation. Animal models that reflect human consumption are critical to study the impact of cannabis on brain and behaviour. The study found different pharmacokinetic profiles following inhalation versus injection of THC, with higher initial brain THC concentration following inhalation.

Oral efficacy of Δ(9)-tetrahydrocannabinol and cannabidiol in a mouse neuropathic pain model

Vanessa A. Mitchell et al.

Summary: The study found that oral THC and CBD, either alone or in combination, have analgesic effects in an animal neuropathic pain model. However, unlike injection studies, the combination of THC:CBD lacks analgesic synergy when delivered orally. Furthermore, both oral THC and THC:CBD combination display a relatively poor therapeutic window, suggesting that CBD may provide a safer but less effective oral treatment for nerve injury-induced neuropathic pain than THC.

Assessment of cognitive and psychomotor impairment, subjective effects, and blood THC concentrations following acute administration of oral and vaporized cannabis

Tory R. Spindle et al.

Summary: This study examined the acute impairing effects of oral and vaporized cannabis on cognitive and psychomotor performance, with high doses resulting in impairment. The DRUID test was found to be more sensitive than other tests. Novel biomarkers of cannabis exposure and/or behavioral tests like DRUID are needed to accurately detect cannabis impairment.

Pharmacokinetic and pharmacodynamic properties of aerosolized (vaped) THC in adolescent male and female rats

C. M. Ruiz et al.

Summary: This study found sex-dependent differences in the pharmacokinetics of THC and its metabolites in adolescent rats after aerosol dosing, as well as dose- and sex-dependent effects on anxiety-like and exploratory behaviors. The results provide a benchmark for dosing adolescent rats with aerosolized THC and understanding the effects of cannabis on the developing brain.

Cannabis constituents interact at the drug efflux pump BCRP to markedly increase plasma cannabidiolic acid concentrations

Lyndsey L. Anderson et al.

Summary: The diverse bioactive molecules in cannabis can interact pharmacologically, leading to the entourage effect. Research shows that the systemic concentrations of cannabinoids can be substantially altered by interactions between cannabis constituents. Ingesting cannabis extracts can enhance plasma CBDA concentrations significantly by inhibiting BCRP transporters in the intestine.
