Osteoblast-Derived Matrix Vesicles Exhibit Exosomal Traits and a Unique Subset of microRNA: Their Caveolae-Dependent Endocytosis Results in Reduced Osteogenic Differentiation

Matrix vesicles promote bone repair after a femoral bone defect in mice

Summary: Matrix vesicles (MtVs), secreted by osteoblasts, have been found to play a role in the regulation of bone cell biology. This study used collagenase-released EVs (CREVs) containing MtVs to promote bone repair in mice with femoral bone defects. The local administration of CREVs significantly enhanced new bone formation and cartilage formation at the damaged site. However, CREVs did not have the same effect on osteogenic differentiation or mineralization of osteoblasts in vitro. These findings suggest that MtVs have potential for bone regeneration.

Anti-inflammatory and immunomodulatory effects of the extracellular vesicles derived from human umbilical cord mesenchymal stem cells on osteoarthritis via M2 macrophages

The results suggest that hUCMSCs-EVs promote the polarization of M2-type macrophages and the expression of IL-10, inhibit inflammation-associated factors in OA chondrocytes caused by IL-1 beta, and alleviate cartilage degradation during OA progression. The enriched miRNAs in hUCMSCs-EVs and the modulation of the PI3K-Akt signaling pathway may contribute to these therapeutic effects.

Summary: Diabetes mellitus is a major cause of blindness and chronic ulcers in the working age population worldwide. Previous studies have found that differentially expressed circular RNAs (circRNAs) are associated with hyperglycemia-induced endothelial cell damage. The transplantation of adipose-derived stem cells (ADSCs) exosomes, which deliver circRNA, has a better therapeutic effect in enhancing wound healing in diabetic mice. This study identified abnormally expressed circRNAs in endothelial progenitor cells (EPCs) under hyperglycemic conditions using high-throughput sequencing. The results showed that circ-Snhg11 expression decreased in EPCs under hyperglycemic conditions, and its overexpression suppressed hyperglycemia-induced endothelial cell damage and M1-like macrophage polarization.

This suggests that miR-133a-3p plays a key role in regulating osteogenic differentiation through ANKRD44.
