Breast cancer brain metastasis: Current evidence and future directions

Hongna Sun et al.

Summary: Breast cancer is the most common cancer in women and brain metastasis has a significant impact on patient survival and quality of life. Triple-negative breast cancer and human epidermal growth factor receptor 2-positive breast cancer are more likely to develop brain metastases. Despite advancements in breast cancer treatment, brain metastases remain the leading cause of morbidity and mortality. Lack of clinical research often leads to exclusion of breast cancer patients with brain metastases from clinical trials. Understanding the molecular mechanisms of breast cancer brain metastasis is crucial for effective treatment.

Immune determinants of the pre-metastatic niche

Laura Patras et al.

Summary: Primary tumors actively induce immunosuppression in the pre-metastatic niche (PMN), the future sites of metastasis, to prepare for disseminated tumor cell arrival. This article discusses recent advances in understanding the tumor immune microenvironment and provides a comprehensive overview of the immune determinants of PMN's spatiotemporal evolution. The authors also outline emerging therapeutic approaches to alter PMN formation and reverse immunosuppression programs.

Emerging Intrinsic Therapeutic Targets for Metastatic Breast Cancer

Jiawei Li et al.

Summary: Breast cancer is the most common cancer worldwide and the leading cause of cancer-related death in women. Although survival rates have improved for early-stage breast cancer patients, those with advanced or metastatic breast cancer still have low survival rates, necessitating the development of new therapies. Mechanistic insights into metastatic breast cancer have provided opportunities for developing novel therapeutic strategies. This review summarizes the emerging therapeutic targets for metastatic breast cancer, including various signaling pathways, growth factor receptors, immune checkpoint proteins, and breast cancer stem cells. Therapeutic drugs targeting these molecules/pathways are either FDA-approved or under clinical trial.

An FDA-Approved Antifungal, Ketoconazole, and Its Novel Derivative Suppress tGLI1-Mediated Breast Cancer Brain Metastasis by Inhibiting the DNA-Binding Activity of Brain Metastasis-Promoting Transcription Factor tGLI1

Daniel Doheny et al.

Summary: Breast cancer is a common and deadly disease, especially when it metastasizes to the brain. This study identifies the truncated glioma-associated oncogene homolog 1 (tGLI1) as a key player in breast cancer brain metastasis. The FDA-approved antifungal drug ketoconazole (KCZ) and its derivative KCZ-7 are found to inhibit tGLI1, reduce the number of breast cancer stem cells, and suppress breast cancer brain metastasis. These findings suggest that tGLI1 can be targeted for the prevention and treatment of breast cancer brain metastasis.
