New Insights into Bitopic Orthosteric/Allosteric Ligands of Cannabinoid Receptor Type 2

Rebecca Ferrisi et al.

Summary: We have developed a new generation of ligands (JR compounds) that target the CB2R. These compounds combine the pharmacophoric portion of CB2R positive allosteric modulator (EC21a) with that of CB2R selective orthosteric agonist (LV62). Among the tested compounds, JR22a showed dualsteric behavior as a CB2R ligand. Computational studies confirmed the binding mode of JR22a at CB2R, and its potential to prevent neuroinflammation was investigated using a human microglial cell model.

Microglial large extracellular vesicles propagate early synaptic dysfunction in Alzheimer's disease

Martina Gabrielli et al.

Summary: The study reveals that microglial extracellular vesicles carrying amyloid-beta can move along axons and spread synaptic dysfunction. Inhibiting the motion of these vesicles restricts the spread of dysfunction, suggesting a potential strategy to delay Alzheimer's disease progression.

Microglial NF-κB drives tau spreading and toxicity in a mouse model of tauopathy

Chao Wang et al.

Summary: The activation of microglial NF-kappa B signaling is crucial for tau spreading and tau-mediated spatial learning and memory deficits in tauopathy mice. Inhibiting NF-kappa B activation reverses tau-associated microglial states and rescues autophagy deficits. This study establishes the role of microglial NF-kappa B signaling in mediating tau spreading and toxicity in tauopathy.

Design, Synthesis, and Biological Activity of New CB2 Receptor Ligands: from Orthosteric and Allosteric Modulators to Dualsteric/Bitopic Ligands

Francesca Gado et al.

Summary: The design and synthesis of dualsteric/bitopic agents that can interact with both orthosteric and allosteric binding sites presents a novel approach in medicinal chemistry. In this study, we designed and characterized the first CB2R heterobivalent bitopic ligands, which displayed selective targeting of CB2R and functional selectivity for the cAMP signaling pathway. Additionally, the most promising bitopic ligand FD-22a showed anti-inflammatory and antinociceptive activity in in vitro and in vivo models, respectively. Computational studies further confirmed the bitopic nature of these compounds.

Microglia and Alzheimer's Disease

Stefania Merighi et al.

Summary: Neuroinflammation may be a crucial avenue for treating and preventing Alzheimer's disease, with microglia playing a key role in the disease process but potentially becoming impaired in later stages. It is important to identify new biomarkers to assess microglial activity and develop novel therapies to restore their physiological function.

Summary: This study focused on the design, synthesis, and evaluation of two new classes of potential dualsteric CB2R ligands. Compound JR64a showed a bias towards G protein activation and high affinity for CB2R, demonstrating great potential for the treatment of neurodegenerative diseases by efficiently inhibiting inflammation.

Trace Amine-Associated Receptor 1 (TAAR1): Molecular and Clinical Insights for the Treatment of Schizophrenia and Related Comorbidities

Pramod C. Nair et al.

Summary: Schizophrenia is a complex and severe mental illness. Current treatments primarily target dopamine neurotransmission and have limited effects on negative symptoms and cognitive impairments. Recent research on trace amine associated receptor-1 (TAAR1) suggests it may be a promising nondopaminergic target for treating schizophrenia and related disorders, with fewer side effects.

Microglial functional alteration and increased diversity in the challenged brain: Insights into novel targets for intervention

Marie-Eve Tremblay

Summary: Microglia are immune cells in the central nervous system that play a crucial role in maintaining CNS health by clearing toxic debris, modulating neuronal circuit wiring, and responding to environmental challenges. Factors such as chronic stress, inadequate diet, infections, and pollution can compromise microglial functions, leading to altered brain connectivity and behavioral deficits. Understanding how environmental challenges affect microglia may provide novel targets for interventions in neurodevelopmental and neuropsychiatric disorders.
