Subunit microparticulate vaccine delivery using microneedles trigger significant SARS-spike-specific humoral and cellular responses in a preclinical murine model

Smital Patil et al.

Summary: The study aimed to evaluate the synergistic effect of a subunit microparticulate vaccine and microneedles assisted vaccine delivery system against a human coronavirus. In vitro tests showed that the vaccine had good immunogenicity, while in vivo tests on mice demonstrated the production of spike GP-specific antibodies and cellular immune response.

Laser-assisted intradermal delivery of a microparticle vaccine for respiratory syncytial virus induces a robust immune response

Ipshita Menon et al.

Summary: Respiratory syncytial virus (RSV) is a significant public health risk for young children. In this study, virus-like particles (VLP) of the RSV fusion (F) protein were used as a vaccine antigen and encapsulated in a microparticle (MP) matrix to enhance antigen presentation. A painless vaccination method using an ablative laser device was employed for intradermal vaccine delivery. Immunization with the adjuvanted vaccine microparticles resulted in a Th1 polarized immune response and significantly reduced lung viral titer.

An Adjuvanted Inactivated SARS-CoV-2 Microparticulate Vaccine Delivered Using Microneedles Induces a Robust Immune Response in Vaccinated Mice

Sharon Vijayanand et al.

Summary: SARS-CoV-2, the causal agent of COVID-19, frequently mutates and reduces vaccine efficacy against variants. An efficient vaccination system is needed to combat these emerging variants. In this study, an adjuvanted inactivated SARS-CoV-2 microparticulate vaccine delivered via transdermal route using dissolving microneedles induced a robust immune response in vaccinated mice.

A dual-delivery platform for vaccination using antigen-loaded nanoparticles in dissolving microneedles

Keegan Braz Gomes et al.

Summary: Effective vaccines delivered via painless methods would revolutionize vaccination. This study developed fast-dissolving microneedles to deliver antigen-loaded polymeric nanoparticles, achieving a dual-delivery platform for transdermal vaccination. The study demonstrated the feasibility of this approach and its potential for transdermal vaccine administration.

A single-shot vaccine approach for the universal influenza A vaccine candidate M2e

Ranmali Kavishna et al.

Summary: Influenza, also known as flu, is a global public health concern and a significant economic burden. Researchers have been pursuing the development of a broadly protective universal flu vaccine, and have now successfully improved the immunogenicity of the M2e vaccine using a targeting strategy. In mouse studies, this strategy has shown promising results in terms of generating a robust protective response. Translation to humans is now an exciting possibility.

Novel microparticulate Zika vaccine induces a significant immune response in a preclinical murine model after intramuscular administration

Akanksha Kale et al.

Summary: This study developed a polymeric vaccine microparticle containing inactivated Zika virus and adjuvants and evaluated its immunogenicity and antigen presentation effects. In vitro experiments showed that the vaccine microparticle with adjuvants induced higher production of nitric oxide and increased autophagy in dendritic cells. In vivo experiments demonstrated that the vaccine microparticle with adjuvants elicited significant antibody responses in mice. This study provided proof-of-concept for a microparticulate Zika vaccine.

Enhanced Immunogenicity of an Influenza Ectodomain Matrix-2 Protein Virus-like Particle (M2e VLP) Using Polymeric Microparticles for Vaccine Delivery

Keegan Braz Gomes et al.

Summary: In this study, the researchers demonstrated that encapsulating a conserved influenza ectodomain matrix-2 protein virus-like particle (M2e5x VLP) into a pre-crosslinked bovine serum albumin (BSA) polymeric matrix enhanced in vitro antigen immunogenicity and in vivo efficacy. The combination of M2e5x VLP microparticles with adjuvant microparticles showed increased expression of antigen-presenting molecules and induced high levels of specific antibodies and T-cell stimulation in mice. This research utilized the advantages of both VLP and polymeric delivery platforms to enhance antigen immunogenicity and adaptive immunity in vivo.

Microneedle Delivery of an Adjuvanted Microparticulate Vaccine Induces High Antibody Levels in Mice Vaccinated against Coronavirus

Sharon Vijayanand et al.

Summary: This 'proof-of-concept' study tested a microparticulate vaccine delivery system and a transdermal vaccine administration strategy using dissolving microneedles. The results showed that the particulate form of the vaccine was more immunogenic than the antigen suspension in vitro, and it induced increased antibody levels in mice following vaccination.

Enhanced Immunogenicity of Adjuvanted Microparticulate HPV16 Vaccines Administered via the Transdermal Route

Trinh Phuong Vo et al.

Summary: This research developed a microparticulate HPV16 vaccine for transdermal administration and evaluated its immunogenicity in a pre-clinical mouse model. The results showed a significant increase in HPV16 IgG antibodies in the microparticle group and expansion of CD4+, CD45R, CD27 and CD62L cell populations in the vaccinated mice group. This suggests the high efficacy of the microparticulate vaccine when administered via transdermal route.
