The coming decade in precision oncology: six riddles

Adam Wahida et al.

Summary: In this Perspective, the authors discuss six key challenges in precision oncology that need to be addressed in order to improve clinical response to molecular targeted therapies. The core principle of precision oncology is that treatment must be personalized, as each cancer has a unique molecular and immune landscape. Choosing the right combination of drugs for each patient and using them at the appropriate stage of the disease is critical to applying precision medicine.

Clinical relevance of tumour-associated macrophages

Mikael J. Pittet et al.

Summary: The interaction of tumour-associated macrophages (TAMs) with cancer and stromal cells in the tumour microenvironment enables and sustains most of the hallmarks of cancer. The authors of this Review examine the diversity of TAMs in various cancer indications, which is being revisited with the advent of single-cell technologies, and discuss the functional roles of different TAM states, the prognostic and predictive value of TAM-related signatures as well as approaches involving TAMs that are currently being or will soon be tested in clinical trials.

Summary: In this study, an efficient next-generation sequencing (NGS) based algorithm was developed and validated for detecting Epstein-Barr virus (EBV) infection in gastric cancer (GC). EBV infection was found to be an independent predictive factor for immune checkpoint blockade (ICB) efficacy in GC, and CTLA-4, tumor mutational burden (TMB), and SMARCA4 mutation were identified as potential predictive biomarkers for ICB efficacy in EBV-associated GC (EBVaGC).

Prognostic and predictive value of a pathomics signature in gastric cancer

Dexin Chen et al.

Summary: In this study, a pathomics signature (PSGC) derived from multiple features of haematoxylin and eosin-stained slides is proposed as an independent predictor of prognosis for gastric cancer patients. The PSGC, when combined with the TNM staging system, significantly improves the accuracy of predicting prognosis. Additionally, satisfactory chemotherapy benefits are observed in patients with a low PSGC, suggesting its potential usefulness as a predictive indicator for decision-making regarding adjuvant chemotherapy.

CD36-mediated metabolic crosstalk between tumor cells and macrophages affects liver metastasis

Ping Yang et al.

Summary: Our study reveals that the scavenger receptor CD36 is upregulated in metastasis-associated macrophages (MAMs) and deletion of CD36 in MAMs attenuates liver metastasis in mice. MAMs contain more lipid droplets and have the unique capability to engulf tumor cell-derived long-chain fatty acids carried by extracellular vesicles. These lipid-enriched vesicles are preferentially partitioned into macrophages via CD36, fueling their tumor-promoting activities and creating a highly immunosuppressive tumor microenvironment (TME) in liver metastasis.

High-throughput single-cell sequencing in cancer research

Qingzhu Jia et al.

Summary: With advances in sequencing and instrument technology, bioinformatics analysis is being applied to large batches of single cells. Single-cell sequencing can be used to evaluate the characteristics of tumors, stromal cells, or immune cells and understand tumor progression, response to environmental changes, complexity of the tumor microenvironment, and interactions between cells. Additionally, single-cell analysis is crucial for exploring disease progression, guiding clinical decision-making, and facilitating precision treatment.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
