Amelioration of Cyclosporine A-Induced Acute Nephrotoxicity by Cordyceps cicadae Mycelia via Mg+2 Reabsorption and the Inhibition of GRP78-IRE1-CHOP Pathway: In Vivo and In Vitro

Zong-Han Wu et al.

Summary: Artificially cultivated mycelia of Cordyceps cicadae (CCM) have been found to protect against cyclosporine A-induced acute nephrotoxicity by reducing blood urea nitrogen levels, alleviating endoplasmic reticulum stress, improving tubular cell damage, and preventing magnesium ion wasting.

Molecular Mechanisms Underlying TNFα-Induced Mitochondrial Biogenesis in Human Airway Smooth Muscle

Debanjali Dasgupta et al.

Summary: Inflammatory cytokines like TNFa promote airway inflammation. Our study explored the mechanism by which TNFa increases mitochondrial biogenesis in human ASM cells, finding that it involves the phosphorylation of CREB and ATF1, which in turn co-activate the expression of PGC1a. We demonstrated that TNFa treatment of hASM cells resulted in increased mitochondrial volume density, biogenesis, and the activation of downstream transcription factors NRF1, NRF2, and TFAM via the pCREB(S133)/pATF1(S63)/PCG1a pathway.

Biologics and airway remodeling in severe asthma

Gilda Varricchi et al.

Summary: Asthma is a chronic inflammatory airway disease that can result in irreversible airflow obstruction. Biologics have been shown to improve airflow obstruction and possibly reverse fixed remodeling. Therefore, analyzing the therapeutic effects of biologics in severe asthma, both early and late, is crucial.

Inflammation-Induced Protein Unfolding in Airway Smooth Muscle Triggers a Homeostatic Response in Mitochondria

Debanjali Dasgupta et al.

Summary: This article proposed a hypothesis on how to mitigate oxidative stress and improve the elasticity of airway smooth muscle (ASM) in response to airway inflammation. Previous studies showed that TNF-α induces hyper-reactivity in ASM, leading to activation of metabolic pathways and ultimately changes in mitochondrial function.

Suppression of Endoplasmic Reticulum Stress by 4-PBA Protects Against Hyperoxia-Induced Acute Lung Injury via Up-Regulating Claudin-4 Expression

Hsin-Ping Pao et al.

Summary: The study revealed that the ER stress inhibitor 4-PBA can protect against hyperoxia-induced acute lung injury by enhancing claudin-4 expression, leading to prolongation of survival in mice, reduction in inflammatory mediators, and protein levels in bronchoalveolar lavage fluid.

TNF alpha induces mitochondrial fragmentation and biogenesis in human airway smooth muscle

Philippe Delmotte et al.

Summary: In hASM of asthmatic patients, mitochondrial volume density is higher and fragmentation is more common, likely influenced by proinflammatory cytokines. TNF alpha exposure leads to increased mitochondrial biogenesis, volume density, and fragmentation, ultimately reducing O-2 consumption rate per mitochondrion. These changes are associated with alterations in expression of key mitochondrial proteins and increased cell proliferation in hASM.
