Colorectal cancer statistics, 2023

Rebecca L. Siegel et al.

Summary: Colorectal cancer is the second leading cause of cancer death in the United States. The incidence of CRC has slowed down, but it is shifting to a younger age, advanced stage, and left colon/rectum. Mortality from CRC has declined overall, but increased in individuals younger than 50 years and Native Americans younger than 65 years. Uncovering the reasons for rising incidence and improving access to screening and treatment are crucial in combating CRC.

A straightforward method to quantify circulating mRNAs as biomarkers of colorectal cancer

Marie Grosgeorges et al.

Summary: Optimizing the biomarker combination in liquid biopsies is important for personalized medicine. A method was developed to purify and quantify circulating cell-free mRNAs in plasma samples using RT-qPCR. Three colorectal cancer biomarkers (B2M, TIMP-1, and CLU) were found to have significantly higher mRNA levels in plasma of metastatic colorectal cancer patients compared to healthy individuals. The analysis of the sum of these mRNA levels showed high discriminatory performance between the two groups.

Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Clinical management of metastatic colorectal cancer in the era of precision medicine

Fortunato Ciardiello et al.

Summary: This review summarizes the progress in the clinical management of patients with metastatic colorectal cancer (mCRC) in the era of precision medicine. Molecular stratification, based on the current treatment algorithm, has been a significant step towards implementing more effective therapeutic approaches. Integrating tumor gene alterations with tumor and microenvironment gene and protein expression profiling, host immune competence, and applying dynamic changes to precision medicine-based continuum of care, could lead to the identification of individual prognostic and predictive parameters for choosing the most appropriate therapeutic program for each patient.

Colorectal Cancer in Inflammatory Bowel Disease: Mechanisms and Management

Summary: Patients with inflammatory bowel disease (IBD) have an increased risk of developing colorectal cancer (CRC). Chronic inflammation plays a key role in the development of neoplastic progression, resulting in dysplastic precursor lesions. Colitis-associated CRC shares molecular similarities with sporadic CRC and the microbiome and host immune system may also play a role. However, further research is needed to understand the specific molecular differences and their role in inflammation-associated cancers.

Influence of Diagnostic Delay on Survival Rates for Patients with Colorectal Cancer

Maria Padilla-Ruiz et al.

Summary: This study aimed to evaluate the impact of diagnostic delay on the overall survival rate of colorectal cancer patients. The findings showed that a diagnostic delay of less than 30 days was associated with shorter survival, while a delay of more than 30 days did not have this effect. Short delay remained predictive of survival regardless of age, BMI, Charlson index, or TNM stage.

Summary: The number of cancer survivors in the United States is increasing due to population growth, aging, and improved detection and treatment. The most prevalent cancers are prostate, skin melanoma, and colon and rectum for males, and breast, uterine corpus, and thyroid for females. Treatment disparities exist, with Black patients being less likely to be diagnosed with early-stage cancer and having lower surgical receipt rates.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
